Cimetidine-sucralfate drug interaction.

To study a possible drug interaction between cimetidine (C) and sucralfate (S) a crossover design was employed using four beagle dogs in each of the three groups: C alone, S alone, C plus S. A one week washout period elapsed between each treatment. C (300 mg, spiked with 3H material 9.9 microCi) was given P.O. by stomach catheter. S (1000 mg) was given as intact tablet. To group 3, C was administered 1 h after S dosing. In presence of S ka of C decreased significantly from 3.99 +/- 0.75 to 2.31 +/- 0.19 (SD) 1/h, and tmax increased significantly from 0.59 +/- 0.12 to 1.00 +/- 0.08 (SD) h. In presence of C Cmax and AUC (0-8) of aluminum increased from 21.38 +/- 4.78 to 29.25 +/- 5.86 (SD) mcg/ml and from 132.3 +/- 40.8 to 168.1 +/- 22.0 (SD) (mcg/ml) h. Absorption of C is delayed in the presence of S. Concomitant administration of C and S results in increased systemic availability of aluminum.
