Influence of nicotine on DNA metabolism.

We investigated the influence of nicotine and its metabolites on semiconservative DNA synthesis, DNA repair processes, and the rejoining of DNA strand breaks. Using a combined treatment with nicotine and gamma irradiation and counting the number of sister chromatid exchanges, we evaluated the action of nicotine on the whole DNA repair process, especially examining the possibility that DNA is damaged by the action of nicotine. All tests were performed on HeLa cells and verified with human lung fibroblasts. Liver microsomes of aroclor-treated rats were used as the metabolizing agent. Our findings showed that nicotine and its metabolites at concentrations below 1 mmol/l are not toxic in the cell lines used and within this subtoxic range they have no effect on DNA metabolism. Neither was DNA repair obstructed after gamma irradiation nor did the number of sister chromatid exchanges increase.

Mechanistic Monte Carlo simulations have proven invaluable in tackling complex challenges in radiobiology, for example for protecting astronauts from solar particle events (SPEs) during deep space missions which remains an underexplored area. In this study, the Geant4-DNA Monte Carlo code was used to assess the DNA damage caused by SPEs and evaluate the protective effectiveness of a multilayer shelter. By examining the February 1956 and October 1989 SPEs-two extreme cases-the results showed that the proposed shelter reduced DNA damage by up to 57.

Hybrid quantum-classical computing algorithms offer significant potential for accelerating the calculation of the electronic structure of strongly correlated molecules. In this work, we present the first quantum simulation of conical intersections (CIs) in a biomolecule, cytosine, using a superconducting quantum computer. We apply the contracted quantum eigensolver (CQE)─with comparisons to conventional variational quantum deflation (VQD)─to compute the near-degenerate ground and excited states associated with the conical intersection, a key feature governing the photostability of DNA and RNA.
