The aim of this study was to develop pellet formulations that could be used to improve the dissolution and bioavailability of a poorly water-soluble model drug, cisapride. Six different types of pellets were prepared by coating sugar spheres in a fluidized bed coater. When the sugar spheres were single layered containing cisapride and solubilizer such as polysorbate 80, the resulting pellets provided an instant release of cisapride in the simulated gastric fluid.

5-Hydroxytryptamine (5-HT) receptor subtypes involved in 5-HT-induced contraction of the chicken gastrointestinal tract were characterized pharmacologically using subtype-selective agonists and antagonists. The proventriculus (area of stomach adjacent to the oesophagus) and ileum are examined. 5-HT applied cumulatively caused sustained contraction of the proventriculus that was not decreased by tetrodotoxin, atropine or l-nitro-arginine methylester (l-NAME).

Objective: This study examined the development of respiration during the preterm and early post-term periods and the effects of other biological and environmental variables, including sleep state, on this development.

Study Design: Weekly 2-hour sleep observations and respiration recordings were obtained on 134 preterm infants from the time they were no longer critical until hospital discharge; a follow-up observation was conducted 1 to 3 months later.

Results: All respiratory variables, except length of respiratory pauses in quiet sleep, decreased although the rate of this decrease slowed after term for four variables.

Salivation therefore plays an important role in defending the esophageal mucosa against refluxed gastric acid and is evoked by cholinergic stimulation. Both nizatidine and cisapride are reported to increase acetylcholine concentrations in the postganglionic cholinergic synapses.
