A number of glycosphingolipids have been subjected to trifluoroacetolysis. Heating the glycosphingolipid at 100 degrees C for 48 h in mixtures of trifluoroacetic acid (TFA) and trifluoroacetic anhydride (TFAA) varying from 1:1 to 1:100 (v/v) resulted in complete cleavage of the glycosidic bond between the carbohydrate and the unsaturated base of the ceramide. The fatty acid(s) linked to the amino group of sphingosine were also released by transamidation. The carbohydrate portion was stable under these conditions due to stabilization of the glycosidic bonds by the electronegative O-trifluoroacetyl groups. The 2-acetamido-2-deoxy-hexose units in the glycosphingolipids were converted into their N-trifluoroacetyl analogues. The N-trifluoroacetyl functions could be removed and the 2-acetamido-2-deoxy-hexoses could be reconstituted by N-acetylation.

Glucanases are widely applied in industrial applications such as brewing, biomass conversion, food, and animal feed. Glucanases catalyze the hydrolysis of glucan to produce the sugar hemiacetal through hydrolytic cleavage of glycosidic bonds. Current study aimed to investigate structural insights of a glucanase from Clostridium perfringens through blind molecular docking, site-specific molecular docking, molecular dynamics (MD) simulation, and binding energy calculation.

The Coca-Cola Company, One Coca-Cola Plaza, Atlanta, Georgia 30313, United States.

The synthesis of radiolabeled [25-C]-siamenoside I following a synthetic route developed using unlabeled materials is disclosed. The synthesis features an early stage labeling of the mogrol's C25 an oxidative cleavage-reconstruction strategy and regioselective glycosylations directed by protecting group manipulations. This route provided access to adequate amounts of [25-C]-siamenoside I for ADME and PK studies.

G-cleave LC3B biosensor: monitoring autophagy and assessing resveratrol's synergistic impact on doxorubicin-induced apoptosis in breast cancer cells.

Autophagy, a crucial process in cancer, is closely intertwined with both tumor progression and drug resistance development. However, existing methods used to assess autophagy activity often pose invasiveness and time-related constraints, limiting their applicability in preclinical drug investigations. In this study, we developed a non-invasive autophagy detection system (NIADS-autophagy, also called G-cleave LC3B biosensor) by integrating a split-luciferase-based biosensor with an LC3B cleavage sequence, which swiftly identified classic autophagic triggers, such as Earle's Balanced Salt Solution and serum deprivation, through protease-mediated degradation pathways.

There are abundant glycosylated substances such as cellulose, hemicellulose, and phytochemical glycosides in plants, which could be converted into functional chemicals such as monosaccharides, oligosaccharides, and bioactive aglycones by cleavage of glycosidic bonds using glycoside hydrolases (GHs). Among those GHs, β-glucosidase and β-xylosidase are the rate-limiting enzymes for degrading cellulose and hemicellulose, respectively, and can convert a variety of glycosylated substances. These two enzymes play important roles in the high value use of plant resources and have great potential applications.
