Clinical Research Centre, Hospital Pulau Pinang, Georgetown, Malaysia; Ministry of Health, Putrajaya, Malaysia.

Background: The prevalence of multidrug-resistant and extensively drug-resistant pathogens has led to increased reliance on broad-spectrum antimicrobials, such as tigecycline. This medicine is commonly used to treat complicated skin and intraabdominal infections as well as community-acquired pneumonia. However, the increasing use of tigecycline has been linked to serious complications, including acute pancreatitis.

Directly Observed Treatment (DOT) involves the administration of medication and in-person monitoring by a healthcare team alongside the patient to improve adherence to tuberculosis (TB) treatment. Cellobiose lipids (CBLs) are a class of glycolipid biosurfactants produced by various fungal strains. These compounds have gained significant interest due to their surface-active and antifungal properties, which are comparable to traditional synthetic surfactants and antimicrobials. Despite their potential applicability in various cosmetic, pharmaceutical, and agricultural formulations, significantly less research has been focused on their production and purification in comparison to other glycolipid biosurfactants, such as mannosylerythritol lipids (MELs) and sophorolipids.

In addressing the formidable challenge posed by methicillin-resistant Staphylococcus aureus (MRSA), this investigation elucidates a novel therapeutic paradigm by specifically targeting the virulence factor sortase A (SrtA) utilizing Tubuloside A (TnA). SrtA plays a critical role in the pathogenicity of MRSA, primarily by anchoring surface proteins to the bacterial cell wall, which is crucial for the bacterium's ability to colonize and infect host tissues. By inhibiting SrtA, TnA offers a novel and distinct strategy compared to traditional antibiotics.
