The in vitro development of bovine corneal opacity induced by two anionic surfactants, sodium decyl sulphate (NaDS) and sodium lauryl sulphate (NaLS), was monitored using a simple, specially constructed instrument, the opacitometer. Results show clearly that NaDS was more potent, and acted more rapidly, than NaLS, an order of potency which has been shown by in vivo ocular irritancy tests with these agents but not in cytotoxicity tests. The measurement of irritant-induced opacity of bovine isolated cornea may therefore offer another approach to the search for viable in vitro alternatives to in vivo ocular irritancy testing.

Purpose: To investigate the effect of Rho-associated protein kinase (ROCK) inhibitor Y27632 on bioenergetic capacity and resilience of corneal endothelial cells (CECs) under metabolic stress.
Methods: Bovine CECs (BCECs) were treated with Y27632 and subjected to bioenergetic profiling using the Seahorse XFp Analyzer. The effects on adenosine triphosphate (ATP) production through oxidative phosphorylation and glycolysis were measured.

Corneal endothelium cells (CECs) regulate corneal hydration between the leaky barrier of the corneal endothelium and the ionic pumps on the surface of CECs. As CECs do not regenerate, loss of CECs leads to poor vision and corneal blindness. Corneal transplant is the only treatment option; however, there is a severe shortage of donor corneas globally.

Interspecies differences in human, pig and sheep corneal thickness may affect the colonization. Currently, there is no research investigating the impact of these differences, along with variable storage and culture conditions on infection in cornea models. These factors could significantly influence utilizing models for drug testing research.

EEG studies have suggested that cattle perceive pain when bled without stunning. The present study on bleeding without stunning, compared cows that had received a local anaesthetic on the site of the bleeding cut (Lurocaine; Luro cows) one hour before bleeding with a 35 cm knife, with cows that had not (saline: Placebo cows). Various physiological indicators potentially related to pain or stress were evaluated.
