Tyrosinase activity in the Ab hamster amelanotic melanoma cells cultured in serum-free Eagle's MEM increased 3 times after 6 h of primary cell culture. This increase was inhibited completely by cycloheximide, while actinomycin D had no effect on this process. After 24 h of culture in MEM with calf serum, further increase of the tyrosinase activity was inhibited by both cycloheximide and actinomycin D. The data presented may indicate that the increase of tyrosinase activity in the primary cell culture of the Ab melanoma is due initially to the unblocking of translation and later to the activation of transcription of the gene controlling the enzyme.

Introduction: Recent findings show that visible light, particularly blue light, stimulates melanogenesis in human skin, though the underlying mechanisms remain debated. This study aimed to determine the cell damage threshold of non-ionizing blue light on keratinocytes while preserving their ability to stimulate melanogenesis.
Methods: Human keratinocytes (N = 3) and melanocytes (N = 3) were isolated from skin samples of varying Fitzpatrick skin phototypes and irradiated with blue light (λpeak = 457 nm) and UVA light (λpeak = 385 nm).

Dynamic high-pressure microfluidization (DHPM) is an emerging treatment technology and has been widely used for the recovery of natural polysaccharides. The aim of the present contribution is to discuss the DHPM-assisted extraction and processing of polysaccharides from some foods and by-products by reviewing the instrument and working principle, procedures, key parameters, and effects of DHPM on the structures, food properties, and bioactivities of resulting polysaccharides. It was found that a DHPM instrument with Z-type chamber is preferable for extracting polysaccharides, and a DHPM with Y-type chamber is applicable for processing polysaccharides.

Epidermal melanocytes form synaptic-like contacts with cutaneous nerve fibers, but the functional outcome of these connections remains elusive. In this pilot study we used our fully humanized re-innervated skin organ culture model to investigate melanocyte-nerve fiber interactions in UV-B-induced melanogenesis. UV-B-irradiation significantly enhanced melanin content and tyrosinase activity in re-innervated skin compared to non-innervated controls, indicating that neuronal presence is essential for exacerbating pigmentation upon UV-B irradiation in long-term culture.
