Naloxone is antagonistic to endogenous opioids. Giving it shows that these opioids act as controlling mechanisms in the secretion of LH and FSH by the hypothalamic, pituitary system and probably of Gn-RH by the hypothalamus. Endogenous opioids and particularly beta endorphin, which is itself under the control of oestradiol, play an inhibitory role on the secretion of Gn-RH and alter the pulsatile mode of Gn-RH secretion. We could not demonstrate PRL action which has been described by other authors.

An alternative approach for the management of acute and chronic pains involves prolonging the half-life of endogenous opiates, such as enkephalins that are released in response to nociceptive stimuli. This can be achieved through the inhibition of enzymatic pathways responsible for the hydrolysis of these peptides, particularly targeting Aminopeptidase N (APN) and Neutral Endopeptidase (NEP). There is clinical concern about the combined use of alcohol and opiates. Several lines of evidence support an interaction between alcohol and the endogenous opioid system. Thus, we hypothesized that ethanol, by causing the release of opioid peptides, may sensitize the system to the action of exogenous opioids such as morphine.
