Identification of strain-specific markers on Neisseria gonorrhoeae that are capable of differentiating gonococci into a large number of distinct classes could facilitate analysis of patterns of gonorrhea transmission and application of gonorrhea control measures. A panel of 12 monoclonal antibodies to gonococcal outer membrane protein IA (PrIA) and IB (PrIB) was used to classify 1,433 strains serologically in a worldwide survey. Eighteen PrIA and 28 PrIB serovars were identified, and a nomenclature is proposed. Gonococcal strains were classified further by auxotyping. Auxotyping and serotyping served to classify the 1,433 isolates into 107 unique auxotype/serovar classes. Dual classification by auxotype and serovar can be used to identify epidemiologically related gonococcal infections in order to test the effectiveness of innovative, focused measures to control gonorrhea.

Background: Performance of a 16S rRNA analysis of the cervicovaginal microbiome of 220 participants recruited into the T Cell Response against Chlamydia (TRAC) cohort between February 2011 and August 2014 in Allegheny County, Pennsylvania USA detected DNA encoding chlamydial 16S rRNA in samples from seven participants whose tests were negative for Chlamydia trachomatis (CT) and DNA encoding gonococcal 16S rRNA from five participants whose tests were negative for Neisseria gonorrhoeae (NG) infection with the Aptima Combo 2 assay (Hologic).
Methods: We used targeted PCR amplification followed by sequencing to characterize the chlamydial 23S rRNA locus and qPCR to detect gonococcal DNA in residual diagnostic swab eluates or DNA used to generate 16S rRNA libraries.
Results: Discrepant specimens that contained chlamydial DNA carried a diagnostic-avoidant, G1523A nucleotide polymorphism in the 23S rRNA locus identical to variants previously detected in Finland, Denmark, and the UK.

The rapid emergence of extensively antimicrobial-resistant strains, including those resistant to all frontline antibiotics, has led to being labelled a priority pathogen by the World Health Organization, highlighting the need for new antimicrobial treatments. Given its absence in humans, targeting cysteine biosynthesis has been identified as a promising avenue for developing new antimicrobials against bacterial pathogens.

Since 2022, an steeply increase has been seen in the number of gonorrhea infections in the Netherlands: in 2023 in the sexual heath centra 31% more cases were found, especially among groups where this is less obvious, such as high educated young women. Gonorrhea infections can be seen with the classic symptoms of an urethritis (the drip). But also asymptomatic or in more severe ways and in different locations.
