Mycoplasma gallisepticum (MG) isolations in live chickens have been made from swabs obtained primarily from the trachea or nasal exudates. As tracheal swabs are often contaminated with feed and because tracheal swabbing may be stressful to the bird, this study was conducted to determine if swabs from the choanal cleft (palatine fissure) would yield MG isolation rates comparable to MG isolation rates of swabs taken from the trachea. Commercial Leghorns from 17 to 22 weeks of age were inoculated via eyedrop with the F strain of MG. Swabs were made from the trachea or the choanal cleft region (palatine fissure) when the chickens were 58 to 63 weeks of age; MG was isolated from 15 of 101 tracheal swab samples and from 51 of 108 choanal cleft swab samples. This study indicates that swabs taken from the choanal cleft region yield higher isolation rates and are more easily obtained than tracheal swabs.

Mass vaccination against infectious laryngotracheitis virus (ILTV) in drinking water can result in variable initial vaccine take. Partial initial vaccine coverage of 20% with an Australian ILT vaccine (A20) previously resulted in significant protection against virulent ILTV challenge. This follow-up study used the international Serva ILT vaccine strain in a factorial design testing four levels of vaccination coverage (0%, 10%, 20%, or 100% of chicks eye-drop vaccinated with the live vaccine at 7 days of age) and three levels of ILTV challenge (no challenge or challenge at 7 or 21 days postvaccination [DPV]).

Broiler chicks' responses following combined IBV live attenuated Massachusetts and 793B strains through gel, spray or oculonasal (ON) vaccination routes were cross-compared. Subsequently, the responses following IBV M41 challenge of the unvaccinated and vaccinated groups were also assessed. Post-vaccination humoral and mucosal immune responses, alongside viral load kinetics in swabs and tissues, were determined using commercial ELISA assays, monoclonal antibody-based IgG and IgA ELISA assays and qRT-PCR respectively.
