Assessing the Risk for Falls in Older Adults After Initiating Gabapentin Versus Duloxetine.

Ann Intern Med

January 2025

Center for Healthcare Delivery Sciences, Division of Pharmacoepidemiology and Pharmacoeconomics, Department of Medicine, Brigham and Women's Hospital and Harvard Medical School, Boston, Massachusetts (R.J.D., N.K.C., N.H., J.C.L.).

Background: The evidence informing the harms of gabapentin use are at risk of bias from comparing users with nonusers.

Objective: To describe the risk for fall-related outcomes in older adults starting treatment with gabapentin versus duloxetine.

Design: New user, active comparator study using a target trial emulation framework.

Background: Providing ongoing support to the increasing number of caregivers as their needs change in the long-term course of dementia is a severe challenge to any health care system. Conversational artificial intelligence (AI) operating 24/7 may help to tackle this problem.

Objective: This study describes the development of a generative AI chatbot-the PDC30 Chatbot-and evaluates its acceptability in a mixed methods study.

PHENYLALANINE AMMONIA-LYASE 2 regulates secondary metabolism and confers manganese tolerance in Stylosanthes guianensis.

Plant Physiol

January 2025

Key Laboratory of Crop Gene Resources and Germplasm Enhancement in Southern China, Ministry of Agriculture and Rual Affairs/Tropical Crops Genetic Resources Institute, Chinese Academy of Tropical Agricultural Sciences, Haikou, 571101, China.

Stylo (Stylosanthes guianensis) is a tropical legume that exhibits considerable tolerance to manganese (Mn) toxicity, which severely constrains plant growth in acidic soils. To elucidate the Mn detoxification mechanisms in stylo, this study investigated the excess Mn-regulated metabolic profile of stylo roots and examined the role of metabolic enzymes in Mn tolerance. Excess Mn triggered oxidative stress in the two stylo genotypes tested.

Objective: To assess the extent to which the concomitant presence of subclinical myocardial injury or stress and diabetes affects the risk of heart failure (HF) subtypes.

Research Design And Methods: The Jackson Heart Study included Black adults, categorized based on diabetes status, high-sensitivity cardiac troponin I (hs-cTnI), and brain natriuretic peptide (BNP) levels. Subclinical myocardial injury was defined as hs-cTnI ≥4 ng/L in women and ≥6 ng/L in men, and subclinical myocardial stress as BNP ≥35 pg/mL.

Circulating proteins may be promising biomarkers or drug targets. Leveraging genome-wide association studies of type 1 diabetes (18,942 cases and 501,638 controls of European ancestry) and circulating protein abundances (10,708 European ancestry individuals), Mendelian randomization analyses were conducted to assess the associations between circulating abundances of 1,560 candidate proteins and the risk of type 1 diabetes, followed by multiple sensitivity and colocalization analyses, horizontal pleiotropy examinations, and replications. Bulk tissue and single-cell gene expression enrichment analyses were performed to explore candidate tissues and cell types for prioritized proteins.
