A new cryptic plasmid, pTA1030 (4.5 megadaltons, copy number 16), was characterized by restriction analysis, together with some other plasmids of Bacillus subtilis.

Microbial source tracking (MST) is a critical tool for identifying sources of human and animal fecal pollution in aquatic environments. To enhance human fecal pollution tracking, this study evaluated the performance characteristics of pBI143, a cryptic plasmid recently identified for potential MST applications. Nucleic acid samples from ten animal species were screened for pBI143, revealing its presence in a small number of pigs, cows, dogs, cats, and flying fox fecal samples.

Foundational techniques in molecular biology-such as cloning genes, tagging biomolecules for purification or identification, and overexpressing recombinant proteins-rely on introducing non-native or synthetic DNA sequences into organisms. These sequences may be recognized by the transcription and translation machinery in their new context in unintended ways. The cryptic gene expression that sometimes results has been shown to produce genetic instability and mask experimental signals.

Avian pathogenic Escherichia coli (APEC) is a significant cause of worldwide morbidity, mortality, and production loss in the poultry industry. Here, we characterized 115 E. coli isolates from avian-diagnosed colibacillosis cases from Georgia, USA in 2022 as part of a year two follow on surveillance using both current and a newly developed serogrouping tool (Klao9-SeroPCR).

The sweetpotato whitefly, Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae), is a polyphagous pest recognized as composed of several cryptic or sibling species. The Middle East-Asia Minor 1 (MEAM1) and the Mediterranean (MED) putative species are invasive and destructive worldwide. The MEAM1 is established throughout the United States, while MED is documented in 27 states.

The transmission of antibiotic-resistance genes, comprising mobilization and relocation events, orchestrates the dissemination of antimicrobial resistance. Inspired by this evolutionarily successful paradigm, we developed ACTIMOT, a CRISPR-Cas9-based approach to unlock the vast chemical diversity concealed within bacterial genomes. ACTIMOT enables the efficient mobilization and relocation of large DNA fragments from the chromosome to replicative plasmids within the same bacterial cell.
