Research has been made on the progesterone-induced effects upon endometrium on impuberal rats (either intact, or castrated, or castrated and adrenalectomized, or hypophysectomized) after an acute or a protracted treatment, which were made every third hour all through the day. It resulted that the hormone basically shows two distinct activities. The first one causes a modest awakening of the anabolical functions both in the lining and in the glandular epithelia, which results in the building up of small amounts of lipid and glycogen. The second one involves the lamina propria as well, and brings about morphological effects which can be superposed on those induced by estrogens. The former seems an autonomous activity and can mainly be found after long-lasting treatments. The latter, on the contrary is influenced by the administration time and can be detected only during a.m. treatments whereas it cannot be detected during p.m. and night treatments. Besides it is influenced by the hypophysal activity and in particular by gonadotropins. The possible mechanism of these influences are discussed.

The role of adrenal androgens as drivers for castration-resistant prostate cancer (CRPC) growth in humans is generally accepted; however, the value of preclinical mouse models of CRPC is debatable, because mouse adrenals do not produce steroids activating the androgen receptor. In this study, we confirmed the expression of enzymes essential for de novo synthesis of androgens in mouse adrenals, with high intratissue concentration of progesterone (P) and moderate levels of androgens, such as androstenedione, testosterone, and dihydrotestosterone, in the adrenal glands of both intact and orchectomized (ORX) mice. ORX alone had no effect on serum P concentration, whereas orchectomized and adrenalectomized (ORX + ADX) resulted in a significant decrease in serum P and in a further reduction in the low levels of serum androgens (androstenedione, testosterone, and dihydrotestosterone), measured by mass spectrometry.

The selective serotonin (5-HT) reuptake inhibitors (SSRIs) are generally used for the treatment of major depressive disorders, and the 5-HT and σ receptors are considered to be targets for treatment of psychiatric disorders. Some SSRIs such as fluvoxamine have agonistic activity towards for the σ receptor, but it is not known whether the effect on the receptor plays a key role in the pharmacological effects. We have recently demonstrated that fluvoxamine shows an anti-anhedonic effect in picrotoxin-induced model of anxiety/depression, while the SSRI paroxetine, which have little affinity for the σ receptor, does not.
