Immunologic characteristics of cells present in rheumatoid synovium from 15 patients were analyzed either "in situ" on frozen sections or after elution. Monoclonal antibodies directed against T cell subpopulation antigens and anti-Ia-like were used. T cells bearing the phenotype T3+, T4+, Ia+ are numerous, and they appeared gathered in lymphoid follicles and in perivascular areas. They are in close relationship with large Ia+ cells probably macrophagic in nature. Numerous dendritic cells as well as intimal cells are also Ia+. Some ratios T8+/T4+ cells are higher than results previously reported. This discrepancy could be related to drugs received by the patients before surgery.

The complex relationship between inflammation, its effects on neuronal excitability and the ensuing plasticity of dorsal root ganglion (DRG) sensory neurons remains to be fully explored. In this study, we have employed a system of experiments assessing the impact of inflammatory conditioned media derived from activated immune cells on the excitability and activity of DRG neurons and how this relates to subsequent growth responses of these cells. We show here that an early phase of increased neuronal activity in response to inflammatory conditioned media is critical for the engagement of plastic processes and that neuronal excitability profiles are linked through time to the structural phenotype of individual neurons.

Interleukin-6 (IL-6) is a relevant cytokine in rheumatoid arthritis (RA) pathogenesis, potentially activating Janus kinases (JAK)-1, -2, and tyrosine kinase 2 (TYK2), and thus, three signal transducer and activator of transcription (STAT)-1, -3 or - 5 pathways. This pilot study aims to explore differences in phosphorylated (p)STAT3 levels among patients with RA, those not classified as RA (nRA), and healthy donors (HD), providing some clues on the relative contribution of each JAK protein to the downstream of the IL-6-induced STAT3 pathway. To investigate the role and mechanism of triptophenolide (TRI) in resisting rheumatoid arthritis (RA). Network pharmacology analysis results suggested that TRI was related to multiple inflammation-related signaling proteins, and possessed the stable structural configuration. In animal experiments, TRI suppressed RA in mice, inhibited tissue inflammation, and improved synovial injury.
