Peanut agglutinin (PNA) is a plant lectin which has been used for the separation of suppressor T cells in mice. The present study was carried out to determine whether PNA would be useful in the isolation of human suppressor T cells. Normal human peripheral blood mononuclear cells (PBMC) were activated by concanavalin A (Con A) and the resultant blastoid cells were then incubated with PNA-coated autologous erythrocytes to induce the rosette formation. The PNA rosette-forming cells (15-18% of Con A-activated PBMC) were separated from non-rosette-forming cells by differential centrifugation. The rosette-forming cells suppressed pokeweed mitogen-induced polyclonal immunoglobulin synthesis by 2- to 5-fold. This degree of suppression was greater than that seen with non-rosette-forming cells or with unseparated Con A-activated cells. Our data indicate that PNA rosette fractionation provides a simple and useful method for the isolation of human suppressor T cells.

R-loops are critical structures that play pivotal roles in regulating genomic stability and modulating gene expression. This study investigates the interactions between the 5-methylcytosine (mC) methyltransferase NOP2/Sun RNA methyltransferase 2 (NSUN2) and R-loops in the transcriptional dynamics and damage repair process of bladder cancer (BCa) cells. We observed markedly elevated levels of R-loops in BCa cells relative to normal urothelial cells.

Phosphatase and Tensin Homologue (PTEN) is one of the most frequently lost tumor suppressors in cancer and the predominant negative regulator of the PI3K/AKT signaling axis. A growing body of evidence has highlighted the loss of PTEN with immuno-modulatory functions including the upregulation of the programmed death ligand-1 (PD-L1), an altered tumor derived secretome that drives an immunosuppressive tumor immune microenvironment (TIME), and resistance to certain immunotherapies. Given their roles in immunosuppression and tumor growth, we examined whether the loss of PTEN would impact the biogenesis, cargo, and function of extracellular vesicles (EVs) in the context of the anti-tumor associated cytokine interferon-γ (IFN-γ).

The ubiquitin proteasome system (UPS), driven by ubiquitin as a degradation signal, eliminates, in a highly specific manner, 'abnormal' proteins and proteins that completed their function. This process involves a hierarchical cascade of E1, E2, and E3 enzymes. The E3 ubiquitin ligases, act as specific receptors that bind their cognate substrates.

MicroRNAs (miRNAs) are non-coding, endogenous small single-stranded RNA molecules involved in post-transcriptional regulation of gene expression. It has been demonstrated that dysregulation of miRNA plays a major role in tumor formation, proliferation, and metastasis. Therefore, the delivery of anti-miRNA oligonucleotides to block the activity of these oncogenic miRNAs is a high-potential anti-cancer therapy approach.
