Substrate kinetics of arginase were investigated in normal and sciatectomized gastrocnemius muscle of frog. Sciatectomized muscle arginase showed 47% elevation in V while Km showed no change. Sciatectomic activation of muscle arginase accounted for the stepped up urea levels which may be of significant use in combating ammonia toxicity and maintaining chemohomeostasis.

Contrary to general expectation, in humans, we have recently shown that after complete conus cauda lesion, the lower motoneuron denervated myofibers may survive several years. In adult rats, the sciatectomized muscle progresses in 4-6 months from severe atrophy to a dystrophic stage and undergoes a dramatic weight loss; during this process, myofiber death/regeneration processes maintain a decreasing population of very small, but vital myofibers. At the same time, in vitro electrophysiologic recordings show that denervated fibers can maintain membrane excitability longer than they can retain contractile properties.

Effect of unilateral (one leg) and bilateral (two legs) sciatectomy was studied on certain serum constituents in the male frog over a period of 3 weeks. The level of creatine kinase, acid and alkaline phosphatases, urea, glucose and proteins increased initially following sciatectomy and decreased subsequently from day 14 onwards, the per cent change being more pronounced in case of bilaterally sciatectomized frogs. The present results indicate an increase in the rate of deamination of proteins in the liver, an impairment in the transportation processes across the cell membrane and an increase in cellular lysosomal activity on sciatectomy suggesting a change either in the amount or in the rate of various enzyme reactions.

Activities of alanine and aspartate aminotransferases are maintained during the first 5 weeks of growth, but decrease subsequently in normal chick gastrocnemii. In sciatectomized muscles, a 5-fold elevation in these enzymes reveals increased utilization of amino acids as a compensatory metabolic support during denervation atrophy.
