Autoimmunity to a neural antigen is a suspected cause of multiple sclerosis (MS), and a candidate autoantigen is myelin basic protein (MBP). Accordingly, saline extracts of brain from patients with MS and other diseases were prepared and the content of immunoglobulin (Ig) determined. Antibody to MBP was measured with a highly-sensitive solid-phase radioimmunoassay using 125I-staphylococcal Protein A. Anti-MBP activity was detected in brain extracts of all eleven MS patients, and in seven out of the eight brain extracts from the patients with other diseases; however the level of anti-MBP activity was significantly higher in the MS extracts (P less than 0.01). Analysis of the MS brain extracts after purification by affinity chromatography columns revealed that the anti-MBP activity was specifically mediated by IgG and resided in the IgG1, IgG2, and/or IgG4 subclasses.

Berberine (BBR) has demonstrated potent anti-inflammatory effects by modulating macrophage polarization. Nevertheless, the precise mechanisms through which berberine regulates post-injury inflammation within the peripheral nerve system remain elusive. This study seeks to elucidate the role of BBR and its underlying mechanisms in inflammation following peripheral nerve injury (PNI).

We aimed to analyze whether EVs carry antibodies against EBV antigens and the possibility that they could serve as diagnostic and disease activity blood biomarkers in RRMS. This was a prospective and observational study including patients with RRMS with active and inactive disease and healthy controls. Blood EVs were isolated by precipitation.

Stroke is one of the leading causes of death and long-term disabilities worldwide, resulting in a debilitating condition occasioned by disturbances in the cerebral vasculature. Primary damage due to metabolic collapse is a quick outcome following stroke, but a multitude of secondary events, including excitotoxicity, inflammatory response, and oxidative stress cause further cell death and functional impairment. In the present work, we investigated whether a primary ischemic damage into the dorsal striatum may cause secondary damage in the circumjacent corpus callosum (CC).

Intake excessive arsenic (As) is related to the occurrence of peripheral neuropathy. However, both the underlying mechanism and the preventive approach remain largely unknown. In the present study, As treatment significantly decreased the mechanical withdrawal threshold and increased the titer of anti-myelin basic protein antibody in rats, accompanied with damaged BNB.

A Coordinated Dance of Effector and Regulatory T Cells that are Reactive to Different Determinants in Autoimmune Response Orchestration.

Laboratory of Immune Regulation, Department of Medicine, UC San Diego School of Medicine, 9500 Gilman Drive, La Jolla, CA 92093.

This article is about my amazing immunological journey for more than a decade with Eli Sercarz as an achaarya, which in Sanskrit means enlightened mentor. My training and routine interactions with colleagues not only at Eli's laboratory but at University of California at Los Angeles, La Jolla Institute for Immunology, Torrey Pines Institute for Molecular Studies, and University of California, San Diego School of Medicine were instrumental in my continued quest to understand how activation of autoimmune inflammatory cells results in pathology and how inflammatory responses are controlled to keep us healthy. I briefly outline different aspects of immune principles related to the immune response to a self-antigen myelin basic protein (MBP) that induces experimental autoimmune encephalomyelitis, a prototype for multiple sclerosis, and how a coordinated interactive network of regulatory T cells (Tregs) CD4+ (CD4 Tregs) and CD8+ (CD8 Tregs) control the anti-MBP response to maintain immune homeostasis.
