The paper describes the authors' experiences with the determination of serum phenylalanine by Hill's modification of the McCaman-Robins fluorimetric method for the determination of serum phenylalanine by automatic amino acid analysers. The method was modified by use of the dipeptide glycyl-leucine, which is 3 times cheaper to produce than leucyl-alanine, the dipeptide originally used. The above method is satisfactory for determining the serum phenylalanine level in normal clinical practice with the normally available, manually operated apparatuses.

While the branched DNA (bDNA) assay is an established bioanalytical method for measurement of lipid nanoparticle (LNP)-encapsulated messenger RNA (mRNA) pharmacokinetic parameters, reverse transcription-quantitative polymerase chain reaction (RT-qPCR) has been considered as an alternative platform. RT-qPCR and bDNA platforms were compared for sensitivity, specificity, correlation, and overall assay performance using serum and tissue samples from 2 nonclinical mouse studies of a therapeutic mRNA candidate, LNP-PAH-mRNA, which encodes for human phenylalanine hydroxylase enzyme. Pharmacokinetic parameter noncompartmental analysis was completed using Phoenix WinNonlin.

This investigation represents a pioneering effort to examine the therapeutic effects of PCB specifically in the context of CFA-induced mice, as well as to elucidate the underlying mechanisms that facilitate such effects. Our study utilized advanced methodologies, namely high-performance liquid chromatography coupled with mass spectrometry (HPLC-MS)-based metabolomics, alongside comprehensive multivariate data analysis, to identify a distinctive metabolic profile associated with acute inflammation. Through our analyses, we discovered that several potential metabolites were significantly implicated in a variety of critical metabolic pathways.

Velvet bean is a native Indonesian legume containing L-dopa, yet it remains underutilized. The aim of this study was to analyze the effects of different types of tempe (soybean, velvet bean, and their combination) on cognitive function, brain histology, dopamine levels, and serum β-amyloid in rats, as well as to identify the parameters most influencing cognitive function, including brain mass and volume, hippocampal neuron count, and dopamine and β-amyloid levels. Background: Traumatic brain injury (TBI) is a major public health concern worldwide, contributing to high rates of injury-related death and disability. Severe traumatic brain injury (sTBI), although it accounts for only 10% of all TBI cases, results in a mortality rate of 30-40% and a significant burden of disability in those that survive. Aim: This study aimed to explore the possible bidirectional interrelations between fructose-induced metabolic syndrome (MS) and apical periodontitis (AP).
Methodology: Twenty-eight male Wistar rats were distributed into four groups (n = 7, per group): Control (C), AP, Fructose Consumption (FRUT) and Fructose Consumption and AP (FRUT+AP). The rats in groups C and AP received filtered water, while those in groups FRUT and FRUT+AP received a 20% fructose solution mixed with water to induce MS.
