Bacterial translocation and gut microbiome imbalance in an experimental infection model of legionellosis in guinea pigs.

BMC Microbiol

March 2025

Department of Pulmonary and Critical Care Medicine, Shengjing Hospital of China Medical University, No. 36 Sanhao Street, Heping District, Shenyang, 110004, China.

Background: Recent studies have shown that in critically ill patients such as those with sepsis and shock, the lung and gut microbiomes undergo profound changes. Legionella pneumophila (Lp) can cause fatal infection, however, such changes have not been investigated in legionellosis. Here, we evaluated the microbiome of the lungs, blood, liver, and small intestine content in Lp-infected guinea pigs.

Background: Pneumococcal pneumonia continues to be a significant global health burden, affecting both children and adults. Traditional diagnostic methods for sputum analysis remain challenging. The objective of this study was twofold: to develop a rapid and easy-to-perform assay for the identification of Streptococcus pneumoniae (Spn) directly in sputum specimens using fluorescence microscopy, and to characterize with high-resolution confocal microscopy the ultrastructure of pneumococci residing in human sputum.

Ethnopharmacological Relevance: Maimendong decoction (MMDD) originates from the ancient Chinese medical text Synopsis of the Golden Chamber and is a well-established remedy for treating lung diseases. It has demonstrated efficacy in the long-term clinical management of idiopathic pulmonary fibrosis (IPF); however, its underlying mechanisms remain unclear.

Aim Of The Study: This study investigates whether MMDD alleviates IPF by reducing type 2 alveolar epithelial cell (AEC2) senescence and enhancing mitochondrial autophagy.

Purpose: Exosomes (Exos) derived from human umbilical cord mesenchymal stem cells (hUC-MSCs) hold great potential for treating bronchopulmonary dysplasia (BPD); however, safety concerns and effects of intranasal administration remain unexplored. This study aimed to explore the safety of hUC-MSCs and Exos and to investigate the efficacy and bio-distribution of repeated intranasal Exos administration in neonatal BPD models.

Methods: Characteristics of hUC-MSCs and Exos were analyzed.
