Activity-Based Bioluminescent Logic-Gate Probe Reveals Crosstalk Between the Inflammatory Tumor Microenvironment and ALDH1A1 in Cancer Cells.

JACS Au

January 2025

Department of Chemistry, Beckman Institute for Advanced Science and Technology, and Cancer Center at Illinois, University of Illinois Urbana-Champaign, Urbana, Illinois 61801, United States.

Cancer cells with high expression of aldehyde dehydrogenase 1A1 (ALDH1A1) are more resistant to chemotherapy, contribute to tumor progression, and are associated with poor clinical outcomes. ALDH1A1 plays a critical role in protecting cells from reactive aldehydes and, in the case of stem cells, regulates their differentiation through the retinoic acid signaling pathway. Despite the importance of this enzyme, methods to study ALDH1A1 high-expressing cancer cells in vivo remain limited.

Long-term screening with serum prostate-specific antigen (PSA) and systematic prostate biopsies can reduce prostate cancer mortality but leads to unacceptable overdiagnosis. Over the past decade, diagnostic methods have improved and the indolent nature of low-grade prostate cancer has been established. These advances now enable more selective detection of potentially lethal prostate cancer.

Diagnostic approach to limbal stem cell deficiency.

Front Ophthalmol (Lausanne)

January 2025

Department of Ophthalmology and Visual Science, University of Chicago, Chicago, IL, United States.

Limbal stem cell deficiency (LSCD) is an important cause of visual and ocular morbidity. Effective diagnosis and management require a thoughtful and comprehensive evaluation of the ocular surface. This review describes the pathogenesis, diagnosis, and grading of LSCD, as well as characteristic findings via slit lamp examination, confocal microscopy, anterior segment optical coherence tomography (AS-OCT), impression cytology, and OCT angiography.

Development and evaluation of patient-centred polygenic risk score reports for glaucoma screening.

BMC Med Genomics

January 2025

Department of Ophthalmology, Flinders Medical and Health Research Institute, Flinders University, Adelaide, SA, Australia.

Background: Polygenic risk scores (PRS), which provide an individual probabilistic estimate of genetic susceptibility to develop a disease, have shown effective risk stratification for glaucoma onset. However, there is limited best practice evidence for reporting PRS and patient-friendly reports for communicating PRS effectively are lacking. Here we developed patient-centred PRS reports for glaucoma screening based on the literature, and evaluated them with participants using a qualitative research approach.

Resistance mechanisms and therapeutic strategies of CDK4 and CDK6 kinase targeting in cancer.

Nat Cancer

January 2025

Department of Oncological Sciences, Precision Immunology Institute, the Tisch Cancer Institute, Icahn School of Medicine at Mount Sinai, New York, NY, USA.

Cyclin-dependent kinases (CDKs) 4 and 6 (CDK4/6) are important regulators of the cell cycle. Selective CDK4/6 small-molecule inhibitors have shown clinical activity in hormonal receptor-positive (HR) metastatic breast cancer, but their effectiveness remains limited in other cancer types. CDK4/6 degradation and improved selectivity across CDK paralogs are approaches that could expand the effectiveness of CDK4/6 targeting.
