G protein-coupled receptors (GPCRs), the largest family of membrane receptors in the mammalian genomes, regulate almost all known physiological processes by transducing numerous extracellular stimuli including almost two-thirds of endogenous hormones and neurotransmitters. The traditional view held that GPCR signaling occurs exclusively at the cell surface, where the receptors bind with the ligands and undergo conformational changes to recruit and activate heterotrimeric G proteins. However, with the application of advanced biochemical and biophysical techniques, this conventional model is challenged by the elucidation of spatiotemporal GPCR activation with the evidence that receptors can signal from subcellular compartments to exhibit various molecular and cellular responses with physiological and pathophysiological relevance.

Background: Recent advancements in novel anti-human epidermal growth factor receptor 2 (HER2) antibody-drug conjugates (ADCs) have highlighted the emerging HER2-low breast cancer subtype with promising therapeutic efficacy. This study aimed to comparatively analyze the metabolic characteristics and prognostic stratification of HER2-low and HER2-zero breast cancer using baseline fluorine-18 fluorodeoxyglucose (F-FDG) positron emission tomography/computed tomography (PET/CT) imaging.

Methods: Consecutive patients with newly diagnosed breast cancer who underwent F-FDG PET/CT prior to therapy in our hospital were retrospectively reviewed.

Objective: To construct polydopamine (PDA)-based nanoparticles (NPs) for combined chemotherapy (CT) and photothermal therapy (PTT) of thyroid tumors by conjugating doxorubicin (DOX) via Schiff base reaction and decorating with RGD peptide.

Methods: PDA NPs were synthesized using dopamine hydrochloride (DA) as the raw material and reacted with DOX-PEG-NH to obtain PDA-DOX NPs. Subsequently, RGD peptide was coupled with PDA-DOX NPs for modification.

Background: The maximum aortic diameter remains the diagnostic criteria and the indicator for prognosis prediction of abdominal aortic aneurysms (AAA). An additional imaging modality is currently needed to help evaluate the prognosis of AAA as well as early detection of AAA formation. This study evaluated the most effective inflammatory markers for AAA using single-cell sequencing and, from these, developed probes to facilitate in vivo multimodal imaging of AAA inflammation.
