The question of strains in AA amyloidosis.

Sci Rep

January 2025

Department of Immunology, Genetics and Pathology, Uppsala University, Rudbeck Laboratory, C11, 75185, Uppsala, Sweden.

The existence of transmissible amyloid fibril strains has long intrigued the scientific community. The strain theory originates from prion disorders, but here, we provide evidence of strains in systemic amyloidosis. Human AA amyloidosis manifests as two distinct clinical phenotypes called common AA and vascular AA.

Myofibrillar Myopathy.

Vet Clin North Am Equine Pract

January 2025

Department of Clinical Sciences, College of Veterinary Medicine and Biomedical Sciences, Colorado State University, Fort Collins, CO, USA. Electronic address:

Myofibrillar myopathy (MFM) is characterized by segmental disarray of myofibrils and ectopic accumulation of a protein called desmin. Previously thought to be a glycogen storage disease, MFM is now recognized as a stand-alone myopathy. Endurance Arabians with MFM usually present with exertional rhabdomyolysis (MFM-ER) at the end of races, elevated serum muscle enzymes, and myoglobinuria.

Developmental endothelial locus-1 as a potential biomarker for the formation and progression of intracranial aneurysm.

Gene

January 2025

Department of Neurology, The First Affiliated Hospital, Sun Yat-sen University, Guangzhou 510000, China; Guangdong Provincial Key Laboratory of Diagnosis and Treatment of Major Neurological Diseases, The First Affiliated Hospital, Sun Yat-sen University, Guangzhou 510000, China; Department of Neurosurgery, Southwest Hospital, Army Medical University, Chongqing 400038, China; Neuroscience and Behavioral Disorders Program, Duke-NUS Medical School, National University of Singapore, Singapore 169857, Singapore. Electronic address:

Background: Intracranial aneurysms (IAs) are pathological dilations occurring at major branch points of cerebral arteries, which can lead to aneurysmal subarachnoid hemorrhage (aSAH) upon rupture. Recent studies have identified developmental endothelial locus-1 (DEL1) as closely associated with IA, though its role remains not fully understood. This study aimed to investigate serum DEL1 level differences in IA patients and explore its function in vascular endothelial cells.

Serum KNG and FVIII may serve as potential biomarkers for depression.

Behav Brain Res

January 2025

Department of Psychiatry, Renmin Hospital of Wuhan University, Wuhan 430060, Hubei, PR China; Department of Psychiatry and Institute of Neuropsychiatry, Renmin Hospital of Wuhan University, Wuhan 430060, Hubei, PR China; Taikang Center for Life and Medical Sciences, Wuhan University, Wuhan, 430071, China. Electronic address:

Background: The global burden of major depressive disorder (MDD) is rising, with current diagnostic methods hindered by significant subjectivity and low inter-rater reliability. Several studies have implied underlying link between coagulation-related proteins, such as kininogen (KNG) and coagulation factor VIII (FVIII), and depressive symptoms, offering new insights into the exploration of depression biomarkers. This study aims to elucidate the roles of KNG and FVIII in depression, potentially providing a foundational basis for biomarker research in this field.

The United States Food and Drug Administration approved the xanomeline-trospium combination in September 2024 for treating schizophrenia, based in part on three double-blind, randomized placebo-controlled trials in adults with schizophrenia experiencing acute psychosis. This random-effects model pairwise meta-analysis of those three trials found that xanomeline-trospium was comparable to placebo in terms of all-cause discontinuation, discontinuation rate due to adverse events, Simpson-Angus Scale score change, Barnes Akathisia Rating Scale score change, body weight change, body mass index change, blood pressure change, serum total cholesterol change, blood glucose change, QTc interval changes, and the incidence of headache, somnolence, insomnia, dizziness, akathisia, agitation, tachycardia, gastroesophageal reflux disease, diarrhea, increased weight, and decreased appetite. However, xanomeline-trospium was associated with a higher incidence of at least one adverse event, dry mouth, hypertension, nausea, vomiting, dyspepsia, and constipation, and increased serum triglyceride compared with placebo.
