Antibiotic resistance caused by Gram-positive bacteria is a growing global human health threat. Selective discrimination and eradication of Gram-positive bacteria and their biofilms is challenging. Therapeutic strategies with multiple modes of action are urgently needed to address the increase in Gram-positive bacteria-resistant nosocomial infections.

Supplements with their own beneficial effect on hosts are consumed by us. N-acetyl cysteine (NAC) and Vitamin C (Vit C) are antioxidants and supplements, consumed for their beneficial properties. The present investigation evaluates the effect of their antioxidant property on antibiotic efficacy against cells from different physiological states, including exponential and stationary-phase, cell aggregates, and stress-induced persister cells.

Mycoplasma pneumonia, caused by Mycoplasma bovis (Mycoplasmopsis bovis; M. bovis), is linked with severe inflammatory reactions in the lungs and can be challenging to treat with antibiotics. Biofilms play a significant role in bacterial persistence and contribute to the development of chronic lesions.

Type 4 pili (T4P) are multifunctional filaments involved in adhesion, surface motility, biofilm formation, and horizontal gene transfer. These extracellular polymers are surface-exposed and, therefore, act as antigens. The human pathogen Neisseria gonorrhoeae uses pilin antigenic variation to escape immune surveillance, yet it is unclear how antigenic variation impacts most other functions of T4P.
