Self-Illuminating In Situ Hydrogel with Immune-Adjuvant Amplify Cerenkov Radiation-Induced Photodynamic Therapy.

Chem Biomed Imaging

April 2024

State Key Laboratory of Natural Medicines, Key Laboratory of Drug Quality Control and Pharmacovigilance, School of Pharmacy, China Pharmaceutical University, Nanjing 210009, China.

Cerenkov radiation-induced photodynamic therapy (CR-induced PDT) has shown the potential to overcome the light penetration limitation in conventional PDT. In addition, the tumor-associated antigens (TAAs) produced by PDT can initiate an antitumor immune process but only show a limited immunotherapeutic effect without the use of immunotherapeutic agents. Herein, a CR-induced PDT hydrogel (R837/Zr-HG-PpIX) has been developed by in situ formation of a hyaluronic acid (HA)-based hydrogel integrated with internal light source Zr, photosensitizer protoporphyrin IX (PpIX), and immune adjuvant imiquimod (R837).

Site-Specific Modification of Native IgGs with Flexible Drug-Load.

Chembiochem

September 2024

Center for Radiopharmaceutical Science, ETH-PSI-USZ, Paul Scherrer Institute, 5232, Villigen-PSI, Switzerland.

Homogeneous, site-specifically conjugated antibodies have shown to result in antibody-drug conjugates (ADCs) with improved efficacy and tolerability compared to stochastically conjugated ADCs. However, precisely controlling the drug load as well as attaching multiple payload moieties to the antibody remains challenging. Here, we demonstrate the simple and direct modification of native IgG-antibodies at the residue glutamine 295 (Q295) without the need for any protein engineering with flexible drug-to-antibody ratios of one or multiple payloads.

Background: Amiodarone is a source of iodine excess that may persist in the body for long time after its withdrawal. The aim of the present analysis was to evaluate the magnitude and long-term time course of 24-h urinary iodine (UI) excretion in patients on antiarrhythmic therapy with amiodarone.

Methods: 24-h UI excretion and thyroid function were evaluated in 67 patients on amiodarone therapy.

Article Synopsis
  • The study investigates the long-term pollution risks of radioactive nuclides cesium (Cs) and strontium (Sr) in the surface soil of a mining city in China, utilizing various assessment models to analyze contamination levels and health risks.
  • Results show that Cs and Sr concentrations are significantly higher in mining areas compared to other regions, but Sr poses little ecological risk and a low non-carcinogenic health risk to humans.
  • The sources of Cs are primarily natural, linked to the soil's parent material, while Sr contamination is largely attributed to mining activities, providing crucial information for pollution management in the area.

Aim: To investigate the metabolism and disposition characteristics of HSK7653 in healthy male Chinese participants.

Methods: A single oral dose of 80 μCi (25 mg) [C]HSK7653 capsules was administered to six healthy participants, and blood, plasma, urine and faeces were collected. Quantitative and qualitative analysis was conducted to investigate the pharmacokinetics, blood-to-plasma ratio, mass balance and metabolism of HSK7653.
