Hydroxyurea (HU) is frequently used in the treatment of various myeloproliferative neoplasms (MPN) where it reduces cell proliferation by impairing DNA synthesis leading to decreased hematopoiesis. Herein we report a case of a 65-year-old female who was diagnosed with Chronic myeloid leukemia and developed severe hemolytic anemia requiring multiple packed red blood cell (RBC) transfusions while being treated with hydroxyurea. The haemolysis persisted until discontinuation of the drug.

Indolent lymphoma: addressing the needs of survivors.

Leuk Lymphoma

January 2025

School of Clinical Sciences at Monash Health, Lymphoma Research Group, Monash University, Clayton, Australia.

Over the past two decades, there has been a continuous improvement in outcome for patients with indolent lymphoma (iNHL) resulting in a gradual accumulation of survivors. While life expectancy in the current era approaches that of the lymphoma-free population, patients continue to experience lifelong complications of the disease and its treatment affecting general health, emotional, psychological and social wellbeing, relationships, employment, finances, and fitness. Contemporary care models while suited to the management of lymphoma are often lacking when it comes to identification and management of these additional needs.

Antigen receptor ITAMs provide tonic signaling by acting as guanine nucleotide exchange factors to directly activate R-RAS2.

The small GTPase R-RAS2 regulates homeostatic proliferation and survival of T and B lymphocytes and, when present in high amounts, drives the development of B cell chronic lymphocytic leukemia. In normal and leukemic lymphocytes, R-RAS2 constitutively binds to antigen receptors through their immunoreceptor tyrosine-based activation motifs (ITAMs) and promotes tonic activation of the phosphatidylinositol 3-kinase (PI3K) signaling pathway. Here, we examined the molecular mechanisms underlying this direct interaction and its consequences for R-RAS2 activity.

The influence of t(v;22) sole, major route ACAs all (+8, n = 14; +Ph, n = 10; +19, n = 1), and -Y sole on progression-free survival. Survival curves are compared with those of patients with the standard t(9;22) translocation. Other ACAs or complex karyotypes did not influence survival.
