Systematic molecular dissection of key intermediates in xanthones biosynthesis in Cudrania tricuspidata and establishment of a viable heterologous expression system.

Plant Physiol Biochem

January 2025

Key Laboratory of Chemical Biology (Ministry of Education), School of Pharmaceutical Sciences, Cheeloo College of Medicine, Shandong University, Jinan, Shandong, 250012, China. Electronic address:

The major phytochemicals in the roots of Cudrania tricuspidata are prenylated xanthones, exhibiting significant structural diversity and bioactive properties, such as anti-inflammatory, antioxidative, and antitumor effects. The biosynthetic pathways of these compounds have not yet been resolved, limiting their production through synthetic biology. In this study, benzoyl-coenzyme A (CoA) ligase (BZL), benzophenone synthase (BPS), and benzophenone 3'-hydroxylase (B3'H) transcripts involved in the biosynthesis of xanthone were cloned and characterized from C.

The chemical investigation of the fruits of Garcinia schomburgkiana growing in Vietnam led to the isolation of a new anofinic acid derivative, 5-hydroxy-8-methoxyanofinic acid (1), a new xanthone, xanthoschome C (2), and a known synthetic phenolic analogue, 4-(2-hydroxybenzyl)-2-(4-hydroxybenzyl) phenol (3), along with seven known xanthones (4-10). The structures of all isolated compounds were determined using spectroscopic techniques (NMR and MS), in conjunction with comparison to existing literature data. All isolated compounds were assessed for their α-glucosidase inhibitory activity and showed significant inhibition, with IC50 values ranging from 12.

Fructose-driven metabolic disorders, such as obesity, non-alcoholic fatty liver disease (NAFLD), dyslipidemia, and type 2 diabetes, are significant global health challenges. Ketohexokinase C (KHK-C), a key enzyme in fructose metabolism, is a promising therapeutic target. α-Mangostin, a naturally occurring prenylated xanthone, has been identified as an effective KHK-C inhibitor, prompting exploration of its analogs for enhanced efficacy.

Aspergixanthones L-T, nine undescribed prenylxanthone derivatives from Aspergillus stellatus.

Phytochemistry

January 2025

Hubei Key Laboratory of Natural Medicinal Chemistry and Resource Evaluation, School of Pharmacy, Tongji Medical College, Huazhong University of Science and Technology, Wuhan, 430030, China. Electronic address:

Nine undescribed prenylxanthone derivatives, aspergixanthones L-T (1-9), were isolated from the fungus Aspergillus stellatus. Compound 4 represents the first propionylated prenylxanthone, while compounds 7-9 are the first examples of prenylxanthones adorned with a 2,3-butanediol group. Their structures with absolute configurations were elucidated based on comprehensive spectroscopic analyses, ECD calculation, single-crystal X-ray diffraction, Mo(OAc) induced ECD experiment, and Mosher's method.
