Immunoglobulins IgG, IgA, and IgM were isolated from porcine serum and milk, and antiserums against the 3 immunoglobulin classes were prepared. Monospecificity of the antiserums for the gamma-, alpha-, and mu-chains was obtained by absorbing them in agarose-linked immunosorbent columns. These immunosorbents were prepared by linking IgG or IgA-IgM to CNBr-activated agarose. Contaminating anti-alpha2-macroglobulin antibodies in the anti-IgA and anti-IgM serums were removed with agarose-linked fetal globulins.

Due to the characteristics of long-term persistence, leading to extensive temporal and spatial distribution of the environmental pollutant dibutyl phthalate (DBP), analyzing of a large number of food and environment samples over a long period and at a high frequency must be necessary. It is of great significance to construct a simple, rapid, sensitive and high-throughput immunoassay method. For the first time, 8 different DBP hapten molecules were designed using computer simulation to provide DBP epitope informations from both conformation and electrostatic aspects for the target molecule.

Previously established World Health Organization (WHO) International Standards (IS) for anti-HPV16 and HPV18 antibodies are used to harmonize results across human papillomavirus (HPV) serology assays. Here, we present an international collaborative study to establish ISs for antibodies against HPV6 (NIBSC code 19/298), HPV11 (20/174), HPV31 (20/176), HPV33 (19/290), HPV45 (20/178), HPV52 (19/296) and HPV58 (19/300). The candidate standards were prepared using sera from naturally infected individuals.

Bispecific antibodies, including bispecific IgG, are emerging as an important new class of antibody therapeutics. As a result, we, as well as others, have developed engineering strategies designed to facilitate the efficient production of bispecific IgG for clinical development. For example, we have extensively used knobs-into-holes (KIH) mutations to facilitate the heterodimerization of antibody heavy chains and more recently Fab mutations to promote cognate heavy/light chain pairing for efficient assembly of bispecific IgG in single host cells.
