Optimizing Surface Maleimide/cRGD Ratios Enhances Targeting Efficiency of cRGD-Functionalized Nanomedicines.

J Am Chem Soc

January 2025

Department of Pharmacy, The First Affiliated Hospital of University of Science and Technology of China (USTC), Laboratory of Precision and Intelligent Chemistry, Department of Polymer Science and Engineering, University of Science and Technology of China, Hefei 230026, Anhui Province, China.

Thiol-maleimide (MI) chemistry is a cornerstone of bioconjugation strategies, particularly in the development of drug delivery systems. The cyclic arginine-glycine-aspartic acid (cRGD) peptide, recognized for its ability to target the integrin αβ, is commonly employed to functionalize maleimide-bearing nanoparticles (NPs) for fabricating cRGD-functionalized nanomedicines. However, the impact of cRGD density on tumor targeting efficiency remains poorly understood.

A primer for Fibroblast Growth Factor 16 (FGF16).

Differentiation

December 2024

University of California, Los Angeles, Department of Molecular, Cell and Developmental Biology, Los Angeles, CA, USA. Electronic address:

During the discovery of the Fibroblast Growth Factor superfamily, scientists were determined to uncover all the genes that encoded FGF proteins. In 1998, FGF16 was discovered with classical cloning techniques in human and rat heart samples. FGF16 loss- and gain-of-function experiments in several organisms demonstrated a conserved function in vertebrates, and as a component of the FGF9 subfamily of ligands (FGF-E/-9/-20), is functionally conserved and sufficient to rescue loss-of-function phenotypes in invertebrates, like C.

Article Synopsis
  • DOCK8 deficiency is a primary immunodeficiency caused by mutations in exon 45 of the DOCK8 gene, leading to symptoms like high serum IgE, eczema, and increased infections.
  • Using CRISPR/Cas9, researchers created a mouse model with the same exon 45 mutation to study its effects on T cell behavior.
  • The study found that the mutation affects T cell homeostasis, disrupts regulatory T cell development, alters glycolysis in CD4 T cells, and promotes a Th2 response upon infection, providing insights for potential treatments.

Protein corona alleviates adverse biological effects of nanoplastics in breast cancer cells.

Nanoscale

September 2024

NanoDelivery Lab, Department of Molecular Medicine, Sapienza University of Rome, Viale Regina Elena 291, 00161 Rome, Italy.

Pollution from micro- and nanoplastics (MNPs) has long been a topic of concern due to its potential impact on human health. MNPs can circulate through human blood and, thus far, have been found in the lungs, spleen, stomach, liver, kidneys and even in the brain, placenta, and breast milk. While data are already available on the adverse biological effects of pristine MNPs ( oxidative stress, inflammation, cytotoxicity, and even cancer induction), no report thus far clarified whether the same effects are modulated by the formation of a protein corona around MNPs.

15-epi-lipoxin A promotes neutrophil exit from exudates for clearance by splenic macrophages.

FASEB J

July 2024

Pulmonary and Critical Care Medicine Division, Department of Medicine, Brigham and Women's Hospital, Harvard Medical School, Boston, Massachusetts, USA.

Specialized proresolving mediators (SPMs) promote local macrophage efferocytosis but excess leukocytes early in inflammation require additional leukocyte clearance mechanism for resolution. Here, neutrophil clearance mechanisms from localized acute inflammation were investigated in mouse dorsal air pouches. 15-HEPE (15-hydroxy-5Z,8Z,11Z,13E,17Z-eicosapentaenoic acid) levels were increased in the exudates.
