A novel disposable amperometric biosensor strip for determination of blood alcohol concentration (BAC) with small volume of sample has been constructed using screen-printed electrodes (SPE), nanocomposite film and alcohol dehydrogenase (ADH). Firstly, the GNPs-MWCNT-Nafion nanocomposite film modified on a working electrode was made of Nafion-117, multi-wall carbon nanotubes (MWCNT) and gold nanoparticles (GNPs). After that Meldola's blue (MB) acted as an electron transfer mediator and the mixed solution of ADH, nicotinamide adenine dinucleotide (NAD(+)) were modified in order on the nanocomposite film.

By combining immunohistochemical technique with microassay methods, we analyzed regional energy metabolism in vulnerable and tolerant areas of gerbil brains during evolution of neuronal damage after bilateral common carotid artery occlusion for 10 min with subsequent reperfusion. Four animals were used for each reperfusion period. Based on the information from the immunohistochemical examination, we dissected out vulnerable and tolerant subregions of the hippocampus, cerebral cortex, and thalamus from freeze-dried 20-microm-thick sections, and measured the levels of creatine phosphate (P-Cr), adenine nucleotides, guanine nucleotides, and purine bodies by HPLC, and the levels of glucose, glycogen, and lactate by an enzyme-immobilized column method.

Much attention has been focused recently on the detection and physical characterization of individual molecules. Using such methods to study the chemical properties, such as reactivity, of single molecules offers the potential to investigate how these might vary from molecule to molecule, and for individual molecules as a function of time. The complex structures of biomolecules such as enzymes make them particularly attractive targets for studying how subtle changes or differences at the molecular level might influence chemical reactivity.

The activity of hypoxanthine phosphoribosyltransferase (HPRT) was determined in the bovine embryo during early embryonic development. Microassay, using [(3)H] hypoxanthine, was improved to measure enzyme activity in the embryonic extract. This activity depended on the reaction time and the concentration of phosphorybosyl pyrophosphate (PRPP) in a reaction.
