Lomitapide, a microsomal triglyceride transfer protein inhibitor, is a lipid-lowering drug that inhibits chylomicron formation in enterocytes and very low-density lipoprotein (VLDL) formation in the liver. Previous studies have shown that very low-density lipoprotein and/or low-density lipoprotein (VLDL/LDL) can deliver certain drugs in addition to lipids. Thus, we hypothesized that serum concentrations of drugs that are more likely to be distributed to VLDL/LDL in the serum (referred to as "VLDL/LDL-philic drugs" in this paper) may be altered by co-administered lomitapide.

Impact of different foot orthoses on gait biomechanics in individuals with chronic metatarsalgia.

Groupe de Recherche sur les Affections Neuromusculosquelettiques, Université du Québec à Trois-Rivières, Canada; Department of Human Kinetics, Université du Québec à Trois-Rivières, Canada; Center for Interdisciplinary Research in Rehabilitation and Social Integration (Cirris), Quebec, QC, Canada.

Background: Foot orthoses (FOs) are commonly prescribed to reduce pain and improve function in individuals with musculoskeletal disorders, including those with chronic metatarsalgia (CM). Reducing the mechanical overload under the metatarsal heads during locomotion is the central point of the treatment for CM. Medially wedged FOs (MWFOs) with a metatarsal pad could further reduce pressure loading under the metatarsal heads and modify foot and ankle biomechanics compared to standard FOs (SFOs).

Background: The removal of preformed antibodies with cleaving enzyme like IdeS (Imlifidase) has demonstrated therapeutic potential in organ transplantation for sensitized recipients. However, preformed xenoreactive antibodies (XAbs) against porcine glycans are predominantly IgM and considered detrimental in pig-to-human xenotransplantation.

Methods: Recombinant IceM, an endopeptidase cleaving IgM, was generated in Escherichia coli.

Purpose: To examine the associations and substitutions of dietary sugars [extrinsic (free) or intrinsic (non-free)] as well as dietary starch and fiber intakes for indices of body fat and cardiometabolic health.

Methods: Dietary intake was assessed at multiple times using multi-day 24-hour recalls over 18-months for indices of body fat (body fat %, waist circumference, BMI, and weight change) (n = 1066) and at baseline and 12 months for cardiometabolic outcomes (LDL, HDL, HbA1c) (n = 736). Bayesian modeling was applied to analyze the probabilistic impact of dietary carbohydrate components using credible intervals for association and substitution analyses with repeated measures random effects modeling.

Monomer compounds from natural products are the major source of active pharmaceutical molecules, which provide great opportunities for discovering of new drugs. However, natural products contain a large number of rather complex compounds. It is difficult to obtain high-purity monomer compounds from complex natural products.
