In solutions of creatine kinase, actin and lactic dehydrogenase oscillations of SH-groups are revealed by three independent methods using three different reagents AgNO3, PCMB, DTNB. The variations of the SH-group titre remain after denaturation of the individual protein solution portions. In the solutions of predenaturated proteins no titre oscillations are observed. Possible cause of the observed oscillations of SH-groups titre may be reversible SH--SS transformation.

Traumatic brain injury (TBI) is a public health problem characterized by a combination of immediate mechanical dysfunction of the brain tissue, and secondary damage. Based on the hypothesis that selected targets, such as Na K-ATPase are involved in the secondary damage after TBI and modulation of this enzyme activity by triterpene 3β, 6β, 16β-trihidroxilup-20 (29)-ene (TTHL) supports the ethnomedical applications of this plant, we decided to investigate whether previous TTHL treatment interrupts the progression of pathophysiology induced by TBI. Statistical analyses revealed that percussion fluid injury (FPI) increased Na,K-ATPase activity in all isoform (α and α) 15 min after neuronal injury.

Oscillations in the level of bovine erythrocyte membrane-SH groups available for DTNB are described. The concentrations of slow- and fast-reactive thiols and the kinetic properties of slow and fast reactions between -SH and DTNB revealed two kinds of cyclic changes. Oscillations of short (about 3 h) and long (about 14 h) duration were studied and the possible implications of these variations in membranes are discussed.

Fatty acid synthetase of chicken liver is rapidly and reversibly inactivated by 5,5'-dithiobis-(2-nitrobenzoic acid) (DTNB) at a rate (k2 = 132 mM-1 S-1 in 3 mM EDTA, 1% (v/v) glycerol, pH 7.0, at 25 degrees C) up to 2200 times higher than the reaction of this reagent with simple thiol compounds. The inactivation is caused by the reaction of the phosphopantetheine SH group, since it is protected competitively by either acetyl- or malonyl-CoA, and since the inactivated enzyme is unreactive with the phosphopantetheine label chloroacetyl-CoA but reactive with the cysteine reagent 1,3-dibromopropanone.
