Density dependence describes the regulation of population growth rate by population density. This process is widely observed in insect populations, including vectors such as mosquitoes and agricultural pests that are targets of genetic biocontrol using gene drive technologies. While there continues to be rapid advancement in gene drive molecular design, most studies prioritise gene drive efficacy over ecology, and the role of density-dependent feedback on gene drives remains neglected.

Background: Mosquitoes are efficient vectors of medically significant flaviviruses and serve as hosts for insect-specific flaviviruses (ISFs). Aedes flavivirus (AEFV) is a classical ISF. Given the increasing discovery of ISFs, it is urgent to evaluate the potential risk of ISFs to human health as well as their impact on the transmission of pathogenic flaviviruses.

Mosquito-borne diseases (MBDs) pose increasing threats under future climate change scenarios and an understanding of mosquito population dynamics is pivotal to predicting future risk of MBDs. Most models that describe mosquito population dynamics often assume that adult life-history is independent of adult age and yet mosquito senescence is known to affect mosquito mortality, fecundity and other key biological traits. Despite this, little is known about the effects of adult age at the level of the mosquito population, especially under varying temperature scenarios.

A dramatic malaria resurgence occurred in areas of Uganda between 2020 and 2022 coincident with the switch to clothianidin-based formulations for indoor residual spraying. During the resurgence, numbers increased but when an alternative insecticide, pirimiphos methyl, was reintroduced in 2023, both malaria cases and mosquito density fell. In this study, we investigated possible causes of the resurgence by assessing; 1) whether sufficient quantities of insecticide were sprayed; 2) the residual insecticide bioefficacy against wild mosquitoes and; 3) the insecticide susceptibility of vector populations using standard test tube assays and wall cone assays.
