The relationship between hearing loss (HL) and Parkinson's disease (PD) remains unclear. Using individual-level and summary-level data from the UK Biobank and the largest genome-wide association studies, we examined this link through observational, Mendelian randomization and genetic pleiotropy analyses. Among 158,229 participants, PD risk rose with HL severity especially in elder and males, and hearing aids significantly reduced PD risk in males.

Reconstruction of Gene Regulatory Networks (GRNs) is essential for understanding gene interactions, their impact on cellular processes, and manifestation of diseases, including drug discovery. Among various mathematical and dynamic models used for GRN reconstruction, S-system model, comprising non-linear differential equations, is widely utilised to capture the behaviour of complex biological systems with non-linear and time-dependent interactions. However, as the network size increases, computational demand for network inference grows due to a greater number of estimation parameters, significantly impacting the performance of optimisation algorithms.

Background: Mate-pair sequencing detects both balanced and unbalanced structural variants (SVs) and simultaneously informs in relation to both genomic location and orientation of SVs for enhanced variant classification and clinical interpretation, while chromosomal microarray analysis (CMA) only reports deletion/duplication. Herein, we evaluated its diagnostic utility in a prospective back-to-back prenatal comparative study with CMA.

Methods: From October 2021 to September 2023, 426 fetuses with ultrasound anomalies were prospectively recruited for mate-pair sequencing and CMA in parallel for prenatal genetic diagnosis.
