Revealing New Analytical Insights into RNA Complexes: Divalent siRNA Characterization by Liquid Chromatography and Mass Spectrometry.

Synthetic Molecule Analytical Chemistry, Genentech Inc., South San Francisco, California 94080, United States.

Accurate characterization of therapeutic RNA, including purity and identity, is critical in drug discovery and development. Here, we utilize denaturing and non-denaturing chromatography for the analysis of ∼25 kDa divalent small interfering RNA (di-siRNA), which comprises a complex 2:1 triplex structure. Ion pair reversed-phase (IPRP) liquid chromatography (LC) experiments with UV absorbance and mass spectrometry (MS) showcase a single denaturing LC method for identity confirmation, impurity profiling, and sequencing with automated MS data interpretation.

This work examines a field theory for directed homopolymers in a good solvent. The field theory is based on a lattice model for single- and double-strand polymers with length variables, direction-dependent pairing energy and interactions. As for the less explicit O(n)-symmetric model, there is a close relation to the conventional one-component branched polymer and the associated Lee-Yang problem.

Non-sticky SiN nanonets for single protein denaturation analysis.

Faraday Discuss

October 2024

The Institute for Advanced Studies (IAS), Wuhan University, Wuhan, 430072, P. R. China.

Proteins play crucial roles in nearly all biological activities, with their functional structures deriving from stable folded conformations. Protein denaturation, induced by chemical and physical agents, is a complex process where proteins lose their stable structures, thereby impairing their biological functions. Characterizing protein denaturation at the single-molecule level remains a significant challenge.

Thermo-rheological properties of xanthan solutions: from shear thinning to elasto-viscoplastic behavior.

Soft Matter

August 2024

The thermo-rheological behavior of xanthan solutions with concentrations spanning a wide range is investigated experimentally. After carefully identifying four distinct regimes of concentration we focused on highly concentrated xanthan solutions. By combining several rheological techniques, it is shown for the first time that such solutions belong to the broad class of elasto-viscoplastic materials by exhibiting both a yield stress and elasticity that manifests around the solid-fluid transition.

Polysaccharides are considered to be ideal green raw materials for enhancing biocompatibility and dispersion effects of nanoparticles. In this study, we coated and dispersed ZnO nanoparticles (NPs) using the denaturation-renaturation process of a triple helix glucan lentinan (LNT), induced by changes in pH value within the reaction system. Various ZnO/LNT composites with different particle sizes and crystal morphologies were prepared and characterized.
