Article Synopsis
  • The Fluorescence Integrated Single-Cell Analysis Script (FISCAS) merges fluorescence microscopy with MALDI mass spectrometry imaging (MALDI-MSI) to enhance single-cell analysis by auto-selecting measurement regions and minimizing off-target data.
  • FISCAS applies MALDI-compatible staining to identify membranes, nuclei, and lipid droplets, which allows for the quick collection of fluorescence data before mass spectrometry measurements on stimulated THP-1 cells at various time points.
  • The results reveal significant molecular diversity among individual cells during differentiation into macrophages, highlighting a more detailed analysis of the differentiation phases and potential lipid markers, which showcases the benefits of combining mass spectrometry with fluorescence data.

Article Synopsis
  • - The study focuses on identifying metabolites in Prunella vulgaris, a plant used for both medicinal and functional food purposes, highlighting the limitations of current analytical methods that can only characterize a few compounds due to low resolution and mass coverage.
  • - Researchers utilized advanced techniques like semi-preparative high-performance liquid chromatography and offline two-dimensional liquid chromatography to analyze a broad range of compounds, achieving high-quality data acquisition through a specific mass spectrometer.
  • - The investigation successfully identified 255 compounds from P. vulgaris, including new discoveries of phenolic acids and O-glycosides, which could enhance the development and quality control of this plant's applications.

Chlorinated paraffins (CPs) are complex mixtures of polychlorinated -alkanes with multiple carbon- (C-, = 9-30) and chlorine homologues (Cl-, = 3-18). The mass spectrometric analysis of CPs is time-consuming and challenging, especially when interferences between CPs, their transformation products, or from the matrix are numerous. These analytical challenges and the lack of appropriate and accessible data evaluation tools are obstacles to their analysis.

Profiling of aberrant sialylated N-glycans in hepatocellular carcinoma by liquid chromatography mass spectrometry.

Clin Chim Acta

March 2024

Department of infectious diseases, Changzheng Hospital, Naval Medical University, Shanghai 200003, China. Electronic address:

Background: Aberrant sialylation is closely associated with the tumorigenesis, progression, and metastasis, and may be of importance for disease diagnosis. However, the analysis of altered expression of sialylated glycans (SGs) in blood is particularly challenging due to the low content and poor ionization efficiency of sialylated glycans in mass spectrometry.

Methods: An analytical strategy based on enrichment of SGs, liquid chromatography-high resolution mass spectrometric detection, and automatic glycan annotation was developed to profile the sialylated N-glycome in serum.
