Toxic Effects of Cobalt on Erythroid Progenitor Cells.

Chem Res Toxicol

Cobalt is a crucial trace element that widely exists in natural environments and is necessary for normal physiological function. However, excessive cobalt exposure leads to various adverse health effects, especially hematological and endocrine dysfunctions. Here, we investigated the toxicity of cobalt on early erythropoiesis by using ex vivo cultured erythroid progenitor cells (EPCs).

Our knowledge of which bone marrow cells affect red cell production is still incomplete. To explore the role of osteocytes in the process we performed bulk RNAseq of osteocytes isolated from control and phlebotomized mice. The top-upregulated gene following phlebotomy was , erythroferrone ( ).

Patients with chronic inflammation are burdened with anemia of inflammation (AI), where inflammatory cytokines inhibit erythropoiesis, impede erythropoietin production, and limit iron availability by inducing the iron regulator hepcidin. High hepcidin hinders iron absorption and recycling, thereby worsening the impaired erythropoiesis by restricting iron availability. AI management is important as anemia impacts quality of life and potentially affects morbidity and mortality.

Objective: Neurocognitive (NC) impairment in people with HIV (PWH) is associated with erythrocyte indices, which may serve as indicators of iron metabolism, inflammation, and related factors. Erythropoiesis requires iron, regulated by a multifaceted system of peptide hormones, including hepcidin. This study postulated that hepcidin might modify the relationship between erythrocyte indices and NC performance in PWH.

PLK1 inhibition impairs erythroid differentiation.

Front Cell Dev Biol

December 2024

School of Life Sciences, Zhengzhou University, Zhengzhou, China.

Polo-like kinase 1 (PLK1), a key regulator of the G2/M phase in mitosis, is frequently overexpressed in numerous tumors. Although PLK1 inhibitors have emerged as promising therapeutic agents for cancer, their use has been linked to significant anemia in a subset of patients, yet the underlying mechanisms remain poorly understood. In this study, we utilized an human umbilical cord blood-derived CD34 cell-based erythroid differentiation system, alongside a murine model, to investigate the impact of PLK1 inhibitors on erythropoiesis.
