Background And Purpose: Parkinson disease (PD) is defined by its unique motor symptoms, where responsiveness to levodopa (L-DOPA) is fundamental for management. Recent research has highlighted a significant relationship between PD symptoms and glymphatic dysfunction. This study endeavors to clarify the connection between glymphatic system functionality and initial motor symptoms in PD, utilizing imaging biomarkers to determine its predictive capacity for L-DOPA responsiveness (LR).

O-GlcNAcylation is a post-translational modification characterized by the covalent attachment of a single moiety of GlcNAc on serine/threonine residues in proteins. Tyrosine hydroxylase (TH), the rate-limiting step enzyme in the catecholamine synthesis pathway and responsible for production of the dopamine precursor, L-DOPA, has its activity regulated by phosphorylation. Here, we show an inverse feedback mechanism between O-GlcNAcylation and phosphorylation of TH at serine 40 (TH pSer40).

Introduction: In 2015, directional leads have been released in Europe for deep brain stimulation (DBS) and have been particularly used for subthalamic nucleus (STN) DBS for Parkinson's disease (PD). In this study we aimed to compare an omnidirectional and directional leads cohort of PD patients when it comes to clinical effectiveness and to assess the correlation with volume of tissue activated - target overlap (VTA-target).
Methods: A total of 60 consecutive patients were retrospectively included.

A scaffold design for tendon regeneration has been proposed, which mimics the microstructural features of tendons and provides appropriate mechanical properties. We synthesized a temperature-triggered shape-memory polymer (SMP) using the ring-opening polymerization of polycaprolactone (PCL) with polyethylene glycol (PEG) as a macroinitiator. We fabricated a micropatterned patch using SMP capillary force lithography, which mimicked a native tendon, for providing physical cues and guiding effects.
