New uses of old drugs hold great promise for clinical translation. Flubendazole, an FDA-approved antiparasitic drug, has been shown to target p53 and promote apoptosis in glioblastoma (GBM) cells. However, its damaging mechanism in GBM remains elusive.

Article Synopsis
  • Tyrosine kinase inhibitors (TKIs) are key treatments for chronic myeloid leukemia (CML), but resistance and intolerance to these drugs pose significant challenges.
  • This research highlights the potential of re-purposed drug mebendazole (MBZ) as an effective anti-cancer agent that can work against both imatinib-sensitive and imatinib-resistant CML cells.
  • MBZ was found to inhibit cell proliferation and induce cell death by targeting BCR/ABL activity, disrupting microtubule formation, and increasing DNA damage, presenting a novel approach for treating TKI-resistant CML patients.

The m6A reader insulin-like growth factor-2 mRNA-binding protein 1 (IGF2BP1) is involved in multiple pathophysiological processes through enhanced expression of the proteins encoded by their target mRNAs. However, the functional role of IGF2BP1-mediated m6A in liver fibrosis remains elusive. Here, we report that IGF2BP1 is highly expressed in activated hepatic stellate cells (HSCs), the major driver of fibrogenesis, and TUBB4B is identified as a potential target of IGF2BP1 by re-analysis of the RNA-seq, RIP-seq, and m6A-seq data.

In this study a cutting-edge approach to producing accurate and computationally efficient interatomic potentials using machine learning algorithms is presented. Specifically, the study focuses on the application of Allegro, a novel machine learning algorithm, running on high-performance GPUs for training potentials. The choice of training parameters plays a pivotal role in the quality of the potential functions.

The cilium is a microtubule-based organelle that plays a pivotal role in embryonic development and maintenance of physiological functions in the human body. In addition to their function as sensors that transduce diverse extracellular signals, including growth factors, fluid flow, and physical forces, cilia are intricately involved in cell cycle regulation and preservation of DNA integrity, as their formation and resorption dynamics are tightly linked to cell cycle progression. Recently, several studies have linked defects in specific ciliary proteins to the DNA damage response.
