In an on-going investigation of bioactive metabolites producing potential endophytic fungi, the strain Lasiodiplodia theobromae (SJF-1) was isolated from a medicinal plant Syzygium cumini. The cultural, morphological and molecular identification was done with the SJF-1 strain. The obtained gene sequence was deposited in NCBI with accession number MG 938644.

Identification of Mellein as a Pathogenic Substance of by UPLC-MS/MS Analysis and Phytotoxic Bioassay.

J Agric Food Chem

August 2021

College of Plant Health and Medicine, Key Laboratory of Integrated Crop Pest Management of Shandong Province, Qingdao Agricultural University, Qingdao 266109, P. R. China.

is a pathogenic fungus that can cause apple ring rot, a destructive apple disease in China. There have been reports on its molecular pathogenesis, but the pathogenic substances still remain unknown. In the present study, instrument analysis including UPLC-high-resolution mass spectrometry (HRMS) and nuclear magnetic resonance showed that fermentation broth contained ()-(-)-mellein, a well-known fungal enantiomer of mellein.

Phytotoxic Metabolites Isolated from the Causal Agent of the Scabby Canker of Cactus Pear ( L.).

Toxins (Basel)

February 2020

Dipartimento Scienze Chimiche, Università di Napoli Federico II, Complesso Universitario Monte S. Angelo, Via Cintia 4, 80126 Napoli, Italy.

Six phytotoxins were obtained from the culture filtrates of the ascomycete , the causal agent of the scabby canker of cactus pear ( L.) in minor Sicily islands. The phytotoxins were identified as (-)-()-mellein (); (±)-botryoisocoumarin A (); (-)-(3,4)- and (-)-(3,4)-4-hydroxymellein ( and ); (-)-terpestacin (); and (+)-3,4-dihydro-4,5,8-trihydroxy-3-methylisocoumarin, which we named (+)-neoisocoumarin ().

A simple, sensitive and reliable quantification and identification method was developed for simultaneous analysis of ochratoxin A (OTA) and its related metabolites ochratoxin alpha (OTα), ochratoxin B (OTB) and mellein. The method was assessed by spiking analytes into blank culture media and dried vine fruits. Performance was tested in terms of accuracy, selectivity and repeatability.

An in planta-expressed polyketide synthase produces (R)-mellein in the wheat pathogen Parastagonospora nodorum.

Appl Environ Microbiol

January 2015

Plant Sciences Division, Research School of Biology, The Australian National University, Canberra, Australian Capital Territory, Australia.

Parastagonospora nodorum is a pathogen of wheat that affects yields globally. Previous transcriptional analysis identified a partially reducing polyketide synthase (PR-PKS) gene, SNOG_00477 (SN477), in P. nodorum that is highly upregulated during infection of wheat leaves.
