Artificial gut Simulator. A scheme to predict intestinal and plasma concentration-time profiles of a weakly basic BCS-II drug, dipyridamole.

Eur J Pharm Biopharm

March 2025

Department of Pharmaceutics, University of Minnesota, Minneapolis, MN 55455, USA; Department of Biomedical Engineering, University of Minnesota, Minneapolis, MN 55455, USA. Electronic address:

The objective of this study was to develop a scheme to predict intestinal and plasma concentration-time profiles of the weakly basic BCS-II drug, Dipyridamole (DPD), using an Artificial Gut Simulator (AGS) integrated with a compartment-based disposition model. In vivo data for this study was obtained from previously published literature. A 3-compartment disposition model was developed using the plasma concentration-time profile of DPD following an intravenous bolus dose.

Plastic additives' environmental impacts remain insufficiently understood due to knowledge gaps in their bioavailability, despite growing concerns from increased plastic use and waste. Additives that are non-covalently bound but strongly interact with polymers can be extractable but not leachable, thus non-bioavailable. Nevertheless, most studies have not distinguished between extractable (EC) and leachable content (LC) in plastic additives.

This study investigates the distinctly different dynamics of atomic carbon and oxygen diffusion, both on the surface and into the bulk of iron multilayer films with face-centered cubic (FCC) (100) and body-centered cubic (BCC) (110) structures, and how these processes impact the recombination behavior of carbon and oxygen, particularly at elevated temperatures. On FCC-iron (γ-iron), CO dissociation occurs around 300 K, leading to the formation of segregated carbide and oxide islands on the surface upon annealing. Above the onset temperature of 600 K, mobile oxygen atoms diffuse to the edge of the carbide islands, where they combine with carbon to form CO.

Glimepiride/hydroxypropyl-β-cyclodextrin inclusion compound: preparation, characterization, and evaluation.

Drug Dev Ind Pharm

March 2025

Key Laboratory for Green Pharmaceutical Technologies and Related Equipment of Ministry of Education, College of Pharmaceutical Sciences, Zhejiang University of Technology, Hangzhou 310014, China.

Objective: To enhance solubility and bioavailability of glimepiride (GM), an inclusion compound of glimepiride/hydroxypropyl-β-cyclodextrin (GM/HP-β-CD) was prepared using mechanical ball milling.

Significance: Based on response surface optimization for the ball milling preparation of the inclusion compound, this study investigates it in vitro and in vivo release and pharmacokinetics.

Methods: GM/HP-β-CD inclusion compound was prepared by optimized ball milling based on response surface methodology and characterized using powder x-ray diffraction, differential scanning calorimetry, scanning electron microscopy and fourier transform infrared spectroscopy, and the stability of the compound were studied.
