Introduction: Wound treatment is a significant health burden in any healthcare system, which requires proper management to minimize pain and prevent bacterial infections that can complicate the wound healing process.
Rationale: There is a need to develop innovative therapies to accelerate wound healing cost-effectively. Herein, two polymer-based nanofibrous systems were developed using poly-lactic-co-glycolic-acid (PLGA) and polyvinylpyrrolidone (PVP) loaded with a combination of an antibiotic (Fusidic acid, FA) and a local anesthetic (Lidocaine, LDC) via electrospinning technique for an expedited healing process by preventing bacterial infections while reducing the pain sensation.

The increasing threat of antimicrobial-resistant bacteria, particularly Staphylococcus aureus, which rapidly develops multidrug resistance and commonly colonizes wound surfaces, demands innovative strategies. Phage-encoded endolysins offer a dual-purpose approach as topical therapies for infectious skin wounds and synergistic agents to reduce high-dose antibiotic dependence. This study explores recombinant CHAPk (rCHAPk), efficiently synthesized within 3 h, displaying broad-spectrum antibacterial activity against 10 Gram-positive strains, including resistant variants, with rapid bactericidal kinetics.

Pregnane X receptor (PXR) is a xenobiotic-sensing nuclear receptor with a well-established role in regulating drug metabolism and clearance. Recent studies have shown that PXR is involved in cell proliferation, apoptosis, immune response, and energy homeostasis. It is important to identify compounds that may modulate PXR activity to prevent drug-drug interactions, distinguish chemicals which could potentially generate toxicity, and identify compounds for further development towards therapeutic usage.

Gram-negative bacteria (GNB) pose a major global public health challenge as they exhibit a remarkable level of resistance to antibiotics. One of the factors responsible for promoting resistance against a wide range of antibiotics is the outer membrane (OM) of Gram-negative bacteria. The OM acts as a barrier that prevents the entry of numerous antibiotics by reducing their influx (due to membrane impermeability) and enhancing their efflux (with the help of efflux pumps).
