Pimozide and Adipic Acid: A New Multicomponent Crystalline Entity for Improved Pharmaceutical Behavior.

Molecules

November 2024

Department of Chemistry, Physical Chemistry Section & C.S.G.I. (Consorzio Interuniversitario per lo Sviluppo dei Sistemi a Grande Interfase), University of Pavia, Via Taramelli 16, 27100 Pavia, Italy.

Pimozide is a first-generation antipsychotic used in the treatment of schizophrenia, Gilles de la Tourette syndrome, and other chronic psychoses. Its in vivo efficacy is limited by poor solubility and consequent poor bioavailability. Therefore, adipic acid was used as a coformer for the preparation of a binary product with improved pharmaceutical properties.

Article Synopsis
  • A systematic review analyzed 10 studies involving 523 patients to evaluate how various antipsychotic medications impact the QTc interval in youngsters.
  • Results indicated that risperidone and aripiprazole had minimal QTc effects, while other drugs like quetiapine and pimozide showed varying levels of prolongation, highlighting a need for more research and careful monitoring when prescribing these medications.

Department of Biology Biotechnology Division, Istanbul University, Istanbul, Turkey.

Lassa fever, caused by the zoonotic Lassa virus (LASV), poses a significant health threat in Africa, leading to thousands of infections and deaths annually and has the potential to spread to other parts of the world. Despite the urgency for effective treatments, there are currently no approved drugs or vaccines for Lassa fever. LASV possesses a unique negative-sense RNA genome, and NP plays a crucial role in viral assembly and infection.

l-3,4-Dihydroxyphenylalanine (levodopa and L-DOPA in this text), alongside dopamine, boasts high biocompatibility, prompting industrial demand for its use as a coating material. Indeed, the effectiveness of L-DOPA is steadily rising as it serves as an oral therapeutic agent for neurodegenerative brain diseases, particularly Parkinson's disease (PD). However, the effects of L-DOPA on the growth and function of astrocytes, the main glial cells, and the most numerous glial cells in the brain, are unknown.

Pimozide-loaded nanostructured lipid carriers: Repurposing strategy against lung cancer.

Sci Prog

November 2024

Pharmacological and Diagnostic Research Center, Faculty of Pharmacy, Al-Ahliyya Amman University, Amman, Jordan.

Article Synopsis
  • The study focused on repurposing pimozide (PMZ) by embedding it in nanostructured lipid carriers (NLC) using a specialized method to enhance drug delivery.
  • The formulated NLCs showed favorable characteristics, including a small particle size, suitable zeta potential, and high encapsulation efficiency, alongside demonstrating pH-sensitive drug release and improved anticancer effects on A549 cell lines.
  • The results indicate that PMZ-NLC could be a promising and effective option for targeted lung cancer therapy, warranting further research into its safety and clinical use.
