Article Synopsis
  • - Aminophylline, a medication often used for severe asthma, can cause arrhythmias, but the exact reasons are not clear
  • - In research, aminophylline significantly inhibited the inward rectifier potassium current in both healthy pig heart cells and failing human heart cells at different concentrations
  • - The study found that aminophylline prolonged action potential duration in porcine heart cells, indicating its potential role in causing arrhythmias, suggesting further exploration of its effects on other ionic currents is necessary

The combination of aminophylline and salbutamol is frequently used in clinical practice in the treatment of obstructive lung diseases. While the side effects (including arrhythmias) of the individual bronchodilator drugs were well described previously, the side effects of combined treatment are almost unknown. We aimed to study the arrhythmogenic potential of combined aminophylline and salbutamol treatment in vitro.

Article Synopsis
  • - The study investigates the effects of aminophylline (AP) on ventilator-induced diaphragmatic dysfunction (VIDD), which increases mortality and morbidity in patients needing mechanical ventilation. It focuses on the involvement of the IGF-1-FOXO1-MURF1 pathway in this dysfunction.
  • - Rat models with VIDD were treated with mechanical ventilation, and various techniques (like lentiviral interference and western blot) were used to analyze protein and mRNA levels, diaphragm contractility, oxidative stress, and mitochondrial stability.
  • - The findings aim to shed light on how AP might ameliorate VIDD and its associated mechanisms, including mitochondrial dynamics and apoptosis, which could lead to improved treatment strategies for patients requiring mechanical

Carvacrol-Loaded Nanoemulsion Promotes Tocolytic and Anti-Dysmenorrhea Effects in Rodents.

Pharmacology

April 2024

Programa de Pós-Graduação em Biociências, Universidade Federal do Vale do São Francisco (UNIVASF), Petrolina,, Brazil.

Introduction: Carvacrol is a phenolic constituent of essential oils that has antinociceptive, anti-inflammatory, and antioxidant activities.

Method: This study aimed to evaluate the in vitro spasmolytic and in vivo anti-dysmenorrhea potential of a nanoemulsion-containing carvacrol (nanoCARV).

Results: In isolated rat uterus, nanoCARV reduced spontaneous contractions (pEC50 = 3.

Asthma is a disease characterized by chronic inflammation and hyper responsiveness of airways. We aimed to assess the relaxant potential of phosphodiesterase-4 (PDE4) inhibitors N-sulfonilhidrazonic derivatives on non-asthmatic and asthmatic guinea pig trachea. Firstly, guinea pigs were sensitized and challenged with ovalbumin, and then morphological, and contractile changes were evaluated resulting from asthma, followed by evaluation of relaxant effect of derivatives on guinea pig trachea and the cAMP levels measurement by ELISA.
