The LIM-domain-only protein LMO2 and its binding partner LDB1 are differentially required for class switch recombination.

Proc Natl Acad Sci U S A

January 2025

Department of Immunology and Microbiology, Key Laboratory of Tropical Disease Control of Ministry of Education, Zhongshan School of Medicine, Sun Yat-sen University, Guangzhou, Guangdong 510000, China.

The LIM-domain-only protein LMO2 interacts with LDB1 in context-dependent multiprotein complexes and plays key roles in erythropoiesis and T cell leukemogenesis, but whether they have any roles in B cells is unclear. Through a CRISPR/Cas9-based loss-of-function screening, we identified LMO2 and LDB1 as factors for class switch recombination (CSR) in murine B cells. LMO2 contributes to CSR at least in part by promoting end joining of DNA double-strand breaks (DSBs) and inhibiting end resection.

Creation and long-term in vitro maintenance of valuable genotype collection is one of the modern approach to conservation of valuable gene pool of woody plants. However, during prolonged cultivation, genetic variability of cells and tissues may accumulate and lead to the loss of valuable characteristics of parental plants. It is therefore important to assess the genetic (including cytogenetic) stability of collection clones.

Background: Three dimensional (3D) cell cultures can be effectively used for drug discovery and development but there are still challenges in their general application to high-throughput screening. In this study, we developed a novel high-throughput chemotherapeutic 3D drug screening system for gastric cancer, named 'Cure-GA', to discover clinically applicable anticancer drugs and predict therapeutic responses.

Methods: Primary cancer cells were isolated from 143 fresh surgical specimens by enzymatic treatment.

In the last thirty years research on relative age effects (RAEs) has exploded in numbers. However, the stability and variability of these effects have hardly been investigated. The three aims of this retrospective study were first to investigate the stability and variability of RAEs over 17 years, second to compare these effects for young female and male athletes, and third to compare these effects between selected and non-selected athletes relative to variability estimates from 17 years prior to assess possible changes in athlete development trends.

This study synthesizes a novel three-dimensional (3D) porous coordination polymer (CP), {[Co(L)₀.₅(H₂O)]·NMP·H₂O} (1), via a solvothermal method in a mixed solvent of water and NMP (1-methyl-2-pyrrolidinone), reacting Co(II) ions with H₄L (1,4-bis(5,6-carboxybenzimidazolylmethyl)benzene). The CP exhibits unique fluorescence properties, emitting at 420 nm under UV light excitation at 350 nm, and serves as a carrier for Mesalazine (MSZ) in therapeutic applications.
