Background: Pharmacoepidemiologic studies assessing drug effectiveness for Alzheimer's disease and related dementias (ADRD) are increasingly popular given the critical need for effective therapies for ADRD. To meet the urgent need for robust dementia ascertainment from real-world data, we aimed to develop a novel algorithm for identifying incident and prevalent dementia in claims.
Method: We developed algorithm candidates by different timing/frequency of dementia diagnosis/treatment to identify dementia from inpatient/outpatient/prescription claims for 6,515 and 3,997 participants from Visits 5 (2011-2013; mean age 75.
Background: Patients with Alzheimer's disease (AD) often experience burdensome neuropsychiatric symptoms, including agitation which occurs in both home and long-term care (LTC) facilities, and is associated with substantial increases in caregiver burden and LTC placements. AXS-05 (45-mg dextromethorphan/105-mg bupropion), a novel, oral NMDA receptor antagonist and sigma-1 receptor agonist, approved by the FDA for major depressive disorder, is being investigated for treatment of AD agitation (ADA). AXS-05 has been evaluated in 2 randomized, double-blind studies: Phase 2 ADVANCE-1 (NCT03226522); Phase 3 ACCORD (NCT04797715).

Method: A randomized, double-blind, placebo-controlled crossover design was used for this trial. Participants with MCI (n = 59) were sorted into one of two sequences: Sham-tVNS or tVNS-Sham.

Background: Aerobic exercise may positively affect brain health, although relationships with cognitive change are mixed. This likely is due to individual differences in the systemic physiological response to exercise. However, the acute effects of exercise on brain metabolism and biomarker responses are not well characterized in older adults or cognitively impaired individuals.

Background: Over the past 3 years, the Global Council on Alzheimer's Disease (GCAD) has conducted research on lived experience and care partner journeys. Specifically, this research has focused on the experiences of individuals from historically underrepresented populations, including LGBTQ+, Black, Hispanic/Latino, and Asian communities. The goal has been to identify how these journeys might diverge across communities, understand various nuances that exist across cultures, and recognize the impact these might have on seeking diagnosis, care, and support.
