AZD1775 synergizes with SLC7A11 inhibition to promote ferroptosis.

Sci China Life Sci

Fudan University Shanghai Cancer Center and Institutes of Biomedical Sciences, Fudan University, Shanghai, 200032, China.

Article Synopsis
  • The study investigates how the tumor suppressor p53 affects the effectiveness of the WEE1 inhibitor AZD1775 in promoting ferroptosis, a type of cell death, in lung cancer cells.
  • AZD1775 inhibits cystine uptake, which is crucial for ferroptosis, and leads to increased expression of SLC7A11, a resistance factor, by altering histone modifications via the WEE1-SETDB1 pathway.
  • Combining AZD1775 with the nucleolar stress inducer Actinomycin D enhances the ability to induce ferroptosis and suppress lung cancer growth, particularly in cells with functional p53.

Background & Aims: Hepatocellular carcinoma (HCC) is one of the leading causes of tumour-related death. Here, we investigated the molecular mechanism of HCC by studying the function of circ_GLIS2.

Methods: Human HCC specimens and cell lines were used.

Background: Hepatitis B Virus (HBV) contributes to liver carcinogenesis via various epigenetic mechanisms. The newly defined epigenetics, epitranscriptomics regulation, has been reported to involve in multiple cancers including Hepatocellular Carcinoma (HCC). Our previous study found that HBx, HBV encodes X protein, mediated H3K4me3 modification in WDR5-dependent manner to involve in HBV infection and contribute to oncogene expression.

Department of hepatobiliary surgery, the First Affiliated Hospital of Hunan Normal University, Hunan Provincial People's Hospital, No.61 Jiefang West Road, Changsha City, Hunan Province 410005, China. Electronic address:

Backgrounds/aims: Hepatocellular carcinoma is recognized as the most common subtype of hepatic cancer. Muskelin 1 antisense RNA (MKLN1-AS) shows prognostic value in hepatitis B virus-hepatocellular carcinoma. The aim of this study is to investigate the detailed biological role of MKLN1-AS and Yes-associated transcriptional regulator 1 (YAP1)-related mechanisms.
