A new 0.01-ml serological microtechnique system produced accurate and reproducible complement-fixation and indirect hemagglutination tests. This equipment involves the use of smaller amounts of antigens and sera, and thus increases the value of the method when employed with expensive reagents or limited qualities of sera.

Seroepidemiology of chlamydial infections of wild ruminants in Spain.

J Wildl Dis

January 2000

Infectious Diseases, Department of Animal Pathology, Faculty of Veterinary Science, Murcia University, Spain.

Chlamydial infections were determined serologically among wild ruminants in the Nature Park of the Sierras de Cazorla, Segura y Las Villas (CNP; Spain). Sampling was done during the period from 1990-95. There were 1,244 blood samples collected, consisting of 490 from fallow deer (Dama dama), 343 from mouflon (Ovis mussimon), 283 from red deer (Cervus elaphus) and 128 from Spanish ibex (Capra pyrenaica).

Subcutaneous and conjunctival vaccination with a live attenuated strain of Salmonella Abortusovis: effect of gestation on serological response of ewes.

Vet Res

June 1995

CNEVA-Sophia-Antipolis, laboratoire de pathologie des petits ruminants et des abeilles, Biot, France.

Annual serological testing of flocks vaccinated by the subcutaneous route with a live, attenuated strain of Salmonella Abortusovis has previously demonstrated the persistence of agglutinating antibodies. It has however been impossible to determine whether the antibodies originated from the vaccination or from an enzootic infection. The serological response, as measured by a microtechnique of seroagglutination using a stained antigen, was studied in an isolated flock of 30 adult ewes.

The aim of the experiment was to evaluate in pregnant ewes the clinical, bacteriological and serological effects of conjunctival instillation of S abortusovis. An individual dose of 1 x 10(10) viable S abortusovis were administered to 20 ewes 92 d after mating. Among 18 pregnant ewes, 8 aborted 110 +/- 13 d after mating, 4 had stillborn lambs and 6 a clinically normal lambing (114 +/- 1.

The Testryp CATT was performed on dried blood samples on filter-paper and on diluted blood using a microtechnique. This method was applied to both sample collection techniques and was evaluated in parallel with the classical Testryp CATT on whole blood, as described in the instructions provided with the reagents by the manufacturer. A total of 2087 people were tested; 453 samples were tested in the laboratory and 1634 during a field survey in 5 villages of a trypanosomiasis focus in Daloa, Côte d'Ivoire.

The complement fixation microtechnique against PHA blasts has been used to study HLA-DQw1, 2, 3 specificities with sera from multiple transfused patients and/or from multiparous women. Several sera (6 or 7) have been used to define each DQ specificity. The sera have been chosen because of their reactivity with cells from HLA-DR 1, 2 or w6 donors (for DQw1), DR3 or 7 donors (for DQw2,) DR4 or 5 donors (for DQw3).
