Reproductive hormones associated with the hypothalamic-pituitary-gonadal (HPG) axis are closely linked to bone homeostasis. In this study, we demonstrate that Gonadotropin inhibitory hormone (GnIH, one of the key reproductive hormones upstream of the HPG axis) plays an indispensable role in regulating bone homeostasis and maintaining bone mass. We find that deficiency of GnIH or its receptor Gpr147 leads to a significant reduction in bone mineral density (BMD) in mice primarily by enhancement of osteoclast activation in vivo and in vitro.

Kisspeptin (Kp), a potent regulator of the hypothalamic-pituitary-gonad axis, was recently shown to be involved in partner preference and sexual receptivity in females. Interestingly, Kp and its receptor (Kiss1r) are expressed in brain regions involved in the reward and motivation of reinforcing behaviors. Therefore, in the present study, we designed 3 experiments to determine the participation of Kp in female sexual behavior and the positive affective (PA) reward state induced by paced mating (PM).

The plains vizcacha is a rodent that shows reactivation of the hypothalamic-pituitary-ovary (HPO) axis activity at mid-gestation. This process is enabled by the secretion of hypothalamic gonadotropin-releasing hormone (GnRH) at mid-gestation, followed by follicle-stimulating hormone (FSH) and luteinizing hormone (LH) secretion. However, a decrease in the pituitary GnRH receptor (GnRHR) expression is concomitantly determined.
