An optical BOD biosensor based on intracellular ATP measurements in genetically modified Saccharomyces cerevisiae.

A biosensor for biochemical oxygen demand (BOD) was developed based on intracellular 5'-adenosine triphosphate (ATP) measurements in Saccharomyces cerevisiae. Intracellular ATP was measured using an engineered protein named ATeam, comprising a bacterial FF-ATP synthase ε subunit sandwiched between cyan fluorescent protein and mVenus, a modified yellow fluorescent protein. Because the binding of ATP to ATeam induces changes in the fluorescence spectra owing to Fӧrster resonance energy transfer, S.

Production of α-ketoisovalerate with whey powder by systemic metabolic engineering of Klebsiella oxytoca.

Microb Cell Fact

October 2024

State Key Laboratory of Microbial Technology, Shandong University, NO.72 Binhai Road, Qingdao, 266237, China.

Article Synopsis
  • Whey is a by-product of the dairy industry with potential pollutant concerns, but it can be fermented to produce valuable chemicals like α-ketoisovalerate.
  • The study used a modified strain of Klebsiella oxytoca (KIV-7) to efficiently produce α-ketoisovalerate by inactivating certain enzymes and overexpressing others, achieving high yields from both lactose and whey powder.
  • The findings suggest that K. oxytoca KIV-7, with integrated genes for α-ketoisovalerate production, can be a stable and effective option for generating this chemical and its derivatives.

Regulation of gene expression in milk secretory pathways during secretory activation in Holstein cows.

J Dairy Sci

December 2024

Division of Animal Sciences, University of Missouri, Columbia, MO 65211. Electronic address:

Increased transcription of genes involved in lactose synthesis is a key feature of secretory activation of the mammary gland. We determined which genes are transcriptionally regulated during the transition from colostrum to milk in cows and whether more frequent milking of one udder half would increase milk yields and alter gene expression. We enrolled 12 Holstein cows immediately after calving, harvested colostrum, then randomly assigned one udder half to 12-h (2X) or 6-h (4X) milking intervals for 48 h after first milking.
