Multiple Myeloma: Genetic and Epigenetic Biomarkers with Clinical Potential.

Int J Mol Sci

December 2024

Laboratory of Molecular Genetics, Department of the Structure and Function of Chromosomes, Institute of Molecular and Cellular Biology, Siberian Branch of the Russian Academy of Sciences, Novosibirsk 630090, Russia.

Multiple myeloma (MM) is characterized by the uncontrolled proliferation of monoclonal plasma cells and accounts for approximately 10% of all hematologic malignancies. The clinical outcomes of MM can exhibit considerable variability. Variability in both the genetic and epigenetic characteristics of MM undeniably contributes to tumor dynamics.

Accurate normalization in miRNA studies requires the use of appropriate endogenous controls, which can vary significantly depending on cell types, treatments, and physiological or pathological conditions. This study aimed to identify suitable endogenous miRNA controls for neural progenitor cells (NPCs) and hippocampal tissues, both of which play crucial roles in neurogenesis. Using small RNA sequencing, we identified the most stable miRNAs in primary mouse NPCs and hippocampal tissues and accessed their stability using NormFinder analysis.

Based on social exchange theory, social psychology theories, and despotic leadership theory, this study explored the impact of petty tyranny on employee turnover intentions. Specifically, the authors examined the mediating effect of toxic workplace environments through emotional exhaustion on this relationship among academicians. The authors surveyed 421 employees using a five-point Likert scale across six universities in Lahore, Pakistan and employed a time-lag research design.

Radiopharmaceutical therapy (RPT) enhances tumor response to immune checkpoint inhibitors (ICI) in preclinical models, but the effects of different radioisotopes have not been thoroughly compared. To evaluate mechanisms of response to RPT+ICI, we used NM600, an alkylphosphocholine selectively taken up by most tumors. Effects of Y-, Lu-, and Ac-NM600 + ICIs were compared in syngeneic murine models, B78 melanoma (poorly immunogenic) and MC38 colorectal cancer (immunogenic).

In this work, a new dual-signal fluorescence strategy based on nano-gold molecular beacon (MB) and in-situ generated silver nano-clusters (NCs) coupled with multiple amplification technique was developed for sensitive detection of miRNA (let-7b). miRNA can recognize both hairpin probe (HP) and auxiliary DNA, inducing dual-cycle amplification-process to release plenty of DNA S2. As the report probe carboxyfluorescein (FAM) was modified on Au nanoparticles (AuNPs), the fluorescent signal was quenched due to the fluorescence resonance energy transfer (FRET).
